Objective:Based on the previous clinical and experimental results, this research dealswith the mechanism that acupuncture therapy of "Tiaoshenyizhi" regulatedapoptosis and HSP70 in brain to supply new experimental proofs for MIDtreated with acupuncture.Method:The rat MID model was made by injecting thrombi into the internal carotidartery. The rats were randomly divided into model group acupuncture group ^non-acupoint group and pseudo-operating group by their weight. Thestimulating points for acupuncture group were "Renzhong' "Neiguan" and"Fengchi", those for non-acupoint group two fixed non-acupoints athypochondria. Both of them were needled one time every day. Model groupand pseudo-operating group were stimulated by grasp at the same time andstrength as acupuncture group and non-acupoint group. After two weeks, allrats were killed by cervical dislocation. The left brains of them were takenout. The apoptosis expression of HSP70 mRNA and its protein in cortex,hippocampus corpus striatum and thalamus were determined by TUNEL, insitu hybridization and immunocytochemical technique with computerizedimage processing. The datum were analyzed with one-way ANOVA bymeans of SPSS 10.0.Results:1 The degree of apoptosis in cortex hippocampus corpus striatum andthalamus in model group was significantly higher than that inpseudo-operating group(P<0.05).2 The degree of apoptosis in cortex hippocampi corpus striatum andthalamus in acupuncture group was significantly lower than that in modelgroup(P<0.05).3 Although the expression of HSP70mRNA and its protein in cortexhippocampus, corpus striatum and thalamus in model group had a tendency toenhance, there was no significant difference(P>0.05) between model andpseudo-operating groups.4 The expression of HSP70mRNA and its protein in cortex hippocampuscorpus striatum and thalamus in acupuncture group was remarkably higherthan that in model group(P<0.05).Conclusion:1 The degree of apoptosis in model group is higher than that inpseudo-operating group. Acupuncture can suppress the apoptosis in in thebrain of rat MID model. There is a special property for acupuncture tosuppress the apoptosis in brain of rat MID model.2 Acupuncture can induce the expression of HSP70 in brian of rat MIDmodel. There is a special property for acupuncture to induce the expression ofHSP70 in brain of rat MID model.
