Objective To investigate the significance of p27, Rb and CyclinDl expression in breast carcinoma as well as the relationship to each other between them. Methods the expressions of p27, Rb and CyclinDl in specimens of 60 cases of breast carcinomas and 20 cases of benign breast tissue were detected by SABC immunohistochemical staining. Results (1) The rate of p27 high expression in breast carcinomas was significantly lower (55%) than that in benign breast tissue(85% P<0. 05). The low expression of p27 in breast carcinomas was markedly related to late TNM stage, high incidence of lymph node metastasis, local recurrence, ER and distant metastasis (P<0.05). (2) The rate of Rb high expression in breast carcinomas was significantly lower (51.7%) than that in benign breast tissue(80% P<0.05). The low expression of Rb in breast carcinomas was markedly associated with late TNM stage, local recurrence and distant metastasis (P<0. 05). (3) The rate of CyclinDl high expression in breast carcinomas was significantly high (63. 3%) than that in benign breast tissue(15% P<0.05). The high expression of CyclinDl in breast carcinomas was highly related with lymph node metastasis, TNM stage, ER, localrecurrence and distant metastasis (P<0. 05). (4) There was a inverse correlation between the expression of p27 and CyclinDl breast carcinoma(P<0.05), while a positive relationship between CyclinDl high expression and Rb low expression (P<0.05). Conclusions Abnormatity of many genes involved in the regulatory pathway of p27, Rb and CyclinDl may be involed in the molecular mechanism of breast cacinomas. The low expression of p27 and RBI and high expression of CyclinDl may play a synergistic role in breast carcinomagenesis.
