Objective To investigate the relationship between HLA-DQA! * 0501 or DQA: * 0201 allele and AITDs in Shandong coastal disricts and to evaluate the effect on the pathogenesis of AITDs.Methods The HLA-DQAi * 0501 and DQAi * 0201 alleles were determined by sequence specific primer-polymerase chain reaction method in 132 AITDs patients,including Graves' disease (GD) 91 cases ( 22 men, 69 women) , Hashimoto's thyroiditis (HT) 41 cases ( 3 men, 38 women) and 54 healthy subjects ( 27 men, 27 women), the difference of their frequencies was analyzed in different groups (Chi square test).Results The frequency of the HLA-DQAj * 0501 allele was significantly higher in GD group than in control (P<0. 05,OR=2. 5), whereas there was no significant difference in HT group (P>0. 05). When the two groups were stratified according to sex, no significant difference was found (P>0. 05). The frequency of the HLA-DQA! * 0201 allele was significantly lower in GD and HT groups than in control (GD P<0. 05,OR= 0. 393; HT P<0. 05,OR = 0. 38, respectively,) Meanwhile the two groups were stratified by sex, the significant difference was found in female patients of two groups compared to female of control. (GD P = 0. 001,OR = 0. 225;HT P<0. 05,OR = 0. 183,respectively). No relationship was found between these genes and GD with positive family history or the ophthalmopathy and HT with positive family history.Conclusion The HLA-DQA1 * 0501 allele maybe was the risk factor for GD patients in Shandong coastal districts, whereas HLA-DQAt * 0201 allele maybe was resistant factor for GD and HT, especially for female patients. HLA-DQAx * 0501 and 0201 alleles had cooperative effect on the pathogenesis of GD and HT.
