Objective To investigate the nutritional status of old people and observe effects of antioxidant micronutrients on cell function , aging and apoptosis.Method Three different ages group health subjects were supplemented with antioxidant micronutrients for 56 days. The subjects were distributed equally for age and sex into 3 groups:group 1:9-lly; group 2:16-24y;group 3:55-86y. In each group subjects were assigned at random to the supplement or placebo groups using double-blind method. Two types of tablets were prepared . One for aged man and youth contains vitamin A 3000IU/d, 3-carotene 1. 5mg/d,witamin E200 mg/d,witamin C800 mg/ d,serium 400mg/d per tablet . The other for children has witamin A 2000IU/d, (3- carotene 1. Omg/d,witamin E100 mg/d,witamin C 500 mg/d,serium 200 mg/d;The tablets for placebo group has the same size and color as ones of supplement group taken without antioxidant micronutrients. Double-blind method was adopted. During experiment period subjects' eating habit didn't change. Blood samples were collected separately at the beginning and 56d. A dietary survy was made on aged man ; Hemoglobin and erythrocyte membrance fluidity were detected , the percentage of peripheral blood lymphocyte apoptosis and the change of apoptosis cell form were demonstrated . Fluorescence polarization method , TUNEL staining method ,light microscope , electron microscope ..fluorescence microscope were adopted during experiment.Results The results of dietary survey in aged people showed that the daily energy intake was adequate , accounting for 87. 35 % RNI. Protein intake accounted for 83. 73%. The intakes of serum, vitamin A ,C and witamin E were 87. 3%, 67. 09%, 58. 28% and 164%. The intakes of calcium and riboflavin were 35. 43% and 43. 9%. Excepting witamin E both of which were significantly lower than RNI. The fluorescence polarization of erythrocyte membrance in PLB of aged adults,youth and children were : 0. 3384,0. 3290,0. 3236 , the difference was cignificant campared with children group (q=5. 15, P<0. 01) showing that erythrocyte membrane fluidity decreases from children , youth to old people. Given antioxidant micronutrients fluorescence polariza-tion of aged man decreased significantly from 0. 3367 to 0. 3016 ; campared with those in placebo group whose fluorescence polarization was 0. 3099, the erythrocyte membrance fluidity increased significantly in supplement group. The percentage of instinctive lymphocyte apoptosis in PLB of aged people increase significantly comparede with children . Antioxidant micronutrients can decrease the percentage of instinctive lymphocyte apoptosis. There is a statistical significant between supplement group and placebo group in aged people (P<0. 05). After supplementing antioxidant micronutrients , the percentage of UV-induced lymphocyte apoptosis is lower significantly than those in placebo group. The result showed that antioxidant micronutrients could improve the ability of injured DNA repairing.The typical changes can be seen under electron microscope . The villi in lymphocyte's surface was disappearing, the cell volume was smaller than nomal and the mitochondrion was swollen. The swelling degree in supplement group was lighter than those in placebo group , in which the shape of mitochondrion was like a ballon. The number of mitochondria increased after supplementation of antioxidant micronutrients .Conclusion The intake of antioxidant micronutrients was lower than PNI except for vitamin E. The intake of calcium and riboflavin is insufficent compared with PNI. Peripheral blood erythrocyte membrance fluidity and cell function decreases gradually with the development of senescence.Erythrocy membrance fluidity was improved after Supplementing antioxidant micronutrients.The percentage of apoptosis in PBL-was increased while the age increasing . Supplementing antioxidant micronutrients can decrease the percentage of instinctive apoptosis and induced-apoptosis in PBLCperipheral blood lymphocyte).
