Objective To observe the effect of insulin-like growth factor binding protein-1 (1G-FBP-1) inhibition to the growth of four types tumors in vitro.Methods Engineering recombinant human IGF-1 (rhIGF-1) or phosphorylated or nonphosphorylated binding protein-1 (pBP-lor npBP-l)that was purified from pregnant woman's amniotic fluid to cultivate 37 cases cancer cells. Then the cultured cells were experimented. MTT observed and determined about IGF-lmRNA's expression by both Digoxin labeled IGF-lcDNA probe's in dot hybridization on nitrocellulose filter.Results Hepatoma and breast tumors , pBP-1 group's cells, their cells' IGF-ImRNA expression were higher than control groupCOPTMD p<0. 01). But IGF-1 and npBP-1 group's cells,its IGF-lmRNA expression was not change.Conclusion The pBP-1 can inhibit the growth of human hepatome or breast cancer cells, and the expression of IGF-lmRNA,which may be regulated and inhibited by pBP-1.
