Objective: The purpose of our testing is to study the effect of TOPO inhibitor on human gastric cancer cells SGC-7901 and to provide important basis for applying topoisomerase inhibitor in treating gastric cancer .Method: SGC-7901 cell line was treated with hydroxycamptothecine (HTCP, TOPO I inhibitor ) or picarcibicin (THP, TOPO II inhibitor ). Cytotoxicity, cell apoptosis and telomerase were evaluated by MTT assay , flow cytometry and TRAP-ELISA respectively .Result :The result showed that HCPT and THP both had remarkable cytotoxic effect on gastric cancer cells SGC-7901. The IC50 of HCPT and THP to SGC-7901 were O.Olmg/ml and 0.2 y g/ml respectively .After exposed to HCPT or THP , SGC-7901 cells presented some morphologic feature of apoptosis .Some typical subdiploid peak phase observed by flow cytometry . The apoptotic rate of SGC-7901 cells were 20.6% and 15.73%. After incubation with O.Olmg/mlHCPT or 0.2 y g/ml THP respectively. Telomerase activity showed a concentration-dependent and time-dependent reduction after the addition of HCPT or THP.Conclusion : Topoisomerase inhibitor HCPT and THPboth can induce apoptosis in gastric cancer SGC-7901 cells and show inhibition result in cell cycle and telomerase activity in vitro. The result provides an experimental basis for HCPT and THP clinical application as TOPO inhibitor.
