Objective: To investigate the anti-proliferation and inducing apoptosis of the Anti-cancer Oral Liquid (ACOL) for MGC-803 gastric cancer cell line in vitro.Methods: Using the MTT assay, cell growth curve, colonogenic assay and sero-pharmacological experiment, the inhibition of cell's growth and the sensitivity of the ACOL for MGC-803 gastric cancer cells was examined and using invert microscope, HE stain, transmission and scanning electron microscope and acridine orange(AO)/ethidium bromide(EB) fluorescence stain, the inhibiting proliferation and inducing apoptosis of ACOL for MGC-803 gastric cancer cells were examined. The expression changes of relating-apoptosis gene proteins (Bcl-2, p53, Fas) were detected by immunohistochemistry (S-P method) before and after treating with the ACOL to explore the possible mechanisms of inhibiting proliferations and inducing apoptosis for MGC-803 cells by treating with the Chinese traditional medicine in vitro.Results: The ACOL significantly inhibited the proliferation of gastric cancercells in vitro and the inhibitive effects were depended on the medicine concentration and treating times. After treating 24 hours on the gastric cancer cells with the 50(g/ml of ACOL, morphologic changes of apoptosis with chromatin margination, karyopyknosis, karyorrhexis, apoptotic body were found by the light and electron microscope. It was showed by using acridine orange (AO) / ethidium bromide (EB) fluorescence staining that MGC-803 gastric cancer cells after treated by the ACOL medicine were not only necrosis but also apoptosis, but the apoptosis was as the dominant way. The Bcl-2,p53 and Fas of relating-apoptosis genes was significantly changed before and after treating with the ACOL(P<0.001).Conclusions: The ACOL has significant inhibition of proliferation and inducing apoptosis for MGC-803 gastric cancer cells in vitro and its possible mechanism of inducing apoptosis for tumor cells might be related with the expression changes of the Bcl-2, p53 and Fas of relating-apoptosis genes. This experiment results will provide theoretical basis for clinical applying and further research of the ACOL.
