Various media and conditions were used to study the effects on mycelia by shaking culture and Ig9(3)4 orthogonal experiment. The result was analysized by direct-viewing analysis method. The magnitude of range indicated that the factors which effected the mycelia growth were carbon source, nitrogen source, KH2P04 and MgSO., in sequence. The result showed that: corn powder was the optimum carbon source and bran is the optimum nitrogen source. The optimum medium was: corn powder 5%, bran 3 % , KH2P04 0. 3 % , MgSO, 0. 15%,VBI 20 u g/lOOml.V^ 30 u g/lOOml. The optimum fermented condition was: culture temperature 26 癈 , speed of revolution 130r/min, pH6.5, the amount of inoculation 15%~20%, time for culturing seed liquid 5 days, the amount of liquid 140ml (500ml triangle flask) , fermenting time 7 days. Under this condition, dried mycelia weight was 1.78g per 100ml fermentend liquid.The proteoglycan in fermented liquid and mycelia were 143. 7mg/100ml and 3750mg/100g (dried mycelia) separately which were extracted with hot water and precipitated with 95% alcohol. The content of polysacchride is 61% in proteoglycan..Antitumor activity of proteoglycan from Phellinus igniarius was studied simultaneously. Mice with Lewis cells and S-180 cells were used for investigation of anti-tumor activity. Proteoglycan from fermated mycelia and liquid of Phellinus igniarius were given by mouth to mice at 12.5,25,50, 100,200mg/kg per day.The results showed that the number of nodes in the lungs and the wet weight of the lungs were reduced remarkably. The survival time of mice was increased as compared to the control group given normal saline by giving 50mg/kg per day of proteoglycan for 2 weeks.But no obvious result can be observed by oral proteoglycan once the tubercle have reached a certain size after implanted, the result also showedthat antitumor activity of proteoglycan from Phellinus igniariushas relation to dose.
