Condylomata acuminata are benign epithelial tumors caused by infection with human papillomaviruses (HPVS) and are characterized by abnormal cell proliferation. Cell cycle is the basic process of cell life activities. At present, more and more scholars conside that after the infection of HPV, the epithelial proliferation has a close relation to cell cycle. The cell cycle modulation in CA is more special and complex than in tumors for the existence of HPV. The runaway cause of the cell cycle in CA can be completely considered to be the influence of HPV to the cell cycle. In cell cycle, the modulation of G1-S transformation is a key point, and Cyclin E, Cyclin Dl and p21WAF1/CIP1 are the main restrict factors in G1 -S. Cyclin E and Cylcin D1 are the positive controlling factors, and p21wAF1/CIP1 is the negative controlling factor . The effect of CyclinE, Cyclin Dl and P21WAF1/CIP1 in CA and their relations to HPV are not clear at present.We used SP immunohistochemistry technique to detect the expression of Cyclin E, Cyclin Dl and P21WAF1/CIP1 in CA,and used in situ hybridization to investigate the expression and distribution of HPV in CA, and analysed the relationship of the four factors.The aim of this study is to clarify the mechanism of the proliferation of CA, and to provide a reliable theoretical basis for the settlement of condylomata acuminata recrudescence.Materials and methods: (1) Sixty five bioptic CA samples and 28 normal men foreskins were collected. All of the tissues were fixed in 10% buffer formalin and embedded in paraffin. (2) In situ hybridization technique was used to examine the expression and distribution of HPV in CA; streptavidin-peroxidase (SP) immunostaning technique was used to detect the expression of CyclinE, CyclmD1 and P21WAF1/CIP1 in all of the tissues.(3)With SAS6.12 software system, x2test and t test were used to analyze the data. There is a statistic significance when P<0.05.Results: (1) After in situ hybridization,all of the 65 CA samples had the positive expression of HPV, but the expression of HPV were negative in all 25 normal men foreskins.(2)The results of immunohistochemisty technique:In CA and normal foreskin tissues, the positive rates of Cyclin E were 64.6%(42/65)and 0%(0/28) respectively. There was a significant difference when the two groups were compared with each other(P<0.01).In CA and normal foreskin tissues, the positive rates of P21WAF1/CIP1 were 67.7%(44/65), 28.6%(8/28) respectively, there was a significant difference as the two groups were compared with each other(P<0.05). In CA and normal foreskin tissues, both the twogroups didn't have positive expression of Cyclin D1. (3) In CA, there were 23 samples which are positive expression of both Cyclin E and p2lWAF1/CIP15 and 2 samples which are negative expression of both Cyclin E and P21WAF1/CIP1. The negative expression of Cyclin E in p21WAF1/CIP1 positive group were 21 samples; and the positive expression of Cyclin E in p21WAF1/CIP1 negative group were 19 samples. In all the CA samples, the consistent expression rate was 38.40 (25/65) ,and the different expression rate was 61.54(40/65). There was a significant negative correlation between the expressions of Cyclin E and P21WAF1/CIP1 (x2=9.072, P>0.05 r=-0.373). (4)In Cyclin E positive expression group and negative expression group,HPV LI were 46.99 + 17.80 and 21.40 + 13.20 respectively, and there was a significant difference between the two groups when comparison was made (PO.01). In p21WAF1/CIP1 positive expression group and negative expression group, HPV LI were 31.27 + 16.61 and 51.91 +20.78 respectively, and there was a significant difference between the two groups(P<0.01).Conclusions: (1) In CA, Cyclin E was highly expressed, and there was a positive relation between the expression of Cyclin E and HPVLI. (2) In CA, P21WAF1/CIP1 was highly expressed and there was a negative relation between the expression of p21WAF1/CIP1 and HPVLI.
