Objective: To investigate the changes of cell cycle-related factors Cyclin E, P21 and E2F-1 in Condyloma Accuminata(CA) and their significance.Methods: HPV-associated Condyloma Accuminata were detected with in situ hybridization, and then the expression and distribution of cell cycle-related factors Cyclin E, P21 and E2F-1 protein in 38 cases formalin-fixed and paraffin-embedded CA and 13 cases normal skin tissues were examined with SP immunohistochemistry technique.Results: 32 of 38 cases CA lesions were positive expression by ISH. The positive staining mainly located in the upper spinous cells with a diffuse intranuclear straining pattern, specially in koilocytes and negative in normal skin. The positive rate of Cyclin E, P21, E2F-1 protein staining in normal skin was 46.2%, 38.4% and 46.2% respectively by immunohistochemistry technique, positive staining focused in the basal cell. The positive rate of CycIinE, P21, E2F-1 protein staining in CA was 84.2%, 86.8% and 81.6% respectively. Compared to normal skin, the expression increased and have significance. The positive singals mainly located in the spinous cells of epidermis, markedly positive in the nuclei of koilocytes and anaplastic cell. The expression of CyclinE protein have correlation with that of P21 protein in CA, neither relationship between E2F-1 protein and Cyclin E protein nor relationship E2F-1 protein and P21 protein.Conclusions: The overexpression of Cyclin E protein might have an important predominant role in the replication process of keratinocytes and human papillomavirus. The high-expression of P21 protein in CA may be a kind of host-celluler defense mechanism against the HPV. While nuclear transcriptor E2F-1 might take effect as an oncogene, and need further studying .The overexpression of Cyclin E, P21and E2F-1 protein in CA induce abnormal proliferation of keratinocytes by coordination or restriction.
