Objective: To investigate the expression of RhoA, Cyclin D1 and P21WAF1/CIP1 protein in breast cancer and its correlation with clincopathologic parameters, and investigate the correlations among RhoA, Cyclin D1 and P21WAF1/CIP1 protein.Methods: RhoA,Cyclin D1 and P21WAF1/CIP1 protein expression were detected by immunohistochemical method in 64 cases of primary breast cancer and 20 cases of normal breast tissue.Results: (1) The positive rates of RhoA, Cyclin D1 and P21WAF1/CIP1 were 5.00%,25.00%,15.00% in normal breast tissue, and 73.44%,59.38%,48.44% in breast cancer tissue. There were significant differences in the positive results of RhoA, Cyclin D1, P21WAF1/CIP1 between normal breast tissue and breast cancer tissue( P < 0.01 ).(2)RhoA protein expression was correlated with histological grade, lymph node metastasis and ER status(P < 0.05), and was not correlated with age and pathologic stage. Cyclin D1 protein expression was correlated with pathologic stage and lymph node metastasis (P < 0.05), and was not correlated with age, histological grade and ER status. P21WAF1/CIP1 protein expression was correlated with pathologic stage, histological grade and lymph node metastasis (P <0.05), and was not correlated with age and ER status.(3) RhoA protein expression was associated negatively with P21WAF1/CIP1 protein(χ~ 2 = 4.548 ,P < 0.05) and was not associated with Cyclin D1 protein.Conclusion : The high expression of RhoA and Cyclin D1 protein,and down-regulation of P21WAF1/CIP1 protein expression are correlated with poor prognosis of breast cancer patients . By suppressing P21WAF1/CIP1 protein, RhoA could regulate cell cycle progression, which is associated with generation, invasion and metastasis of breast cancer.
