The Studies on the Regulation of E-selectin Expression on Vascular Endothelial CellsE-selectin is expressed only on activated vascular endothelial cells. It plays very important roles in many physiological and pathological processes, including inflammation. The studies on the regulation of E-selectin expression on vascular endothelial cells are useful to prevent, diagnosis and treat diseases .The method for inducing E-selectin expression on human umbilical vein endothelial cells (HUVEC) with TNF-alpha was established and standardized. It represents the early phase of acute inflammation on big vessels. We also prepared and identified a monoclonal antibody (McAb) against HUVEC, SZ-106. The heavy chain of the McAb belongs to IgGl subclass. Western blot assay demonstrated that the McAb recognized 25KD protein polypeptide. After HUVEC incubated with Ferulic Acid (FA), a kind of traditional Chinese medicine, SZ-106, SZ-21and SZ-51 (McAbs prepared by our lab) respectively, the expression of E-selectin was reduced by FA but no influence with the McAbs. In order to identify SZ-106, we prepared the extracellular matrix (ECM). By comparing the adhesive rate of endothelial cells between existing matrix and without matrix, we found that the adhesive rate was obviously increased by existing matrix. The effects of FA and SZ-106, SZ-21 on the HUVEC- ECM adhesion were determined by measuring the light density of dyed cells at 570nm by Rose Bengal. It shown that only SZ-21 inhibited the HUVEC- ECM adhesion significantly.
