Objective: To explore levels of serum soluble CD44v6(sCD44v6) and hyaluronic acid (HA) in patients with oral and maxillofacial malignancy and to investigate the correlation between the levels of serum sCD44v6 and HA in patients and the type and stage of oral and maxillofacial malignancy. To investigate the fluctuation of serum sCD44v6 and HA concentration in patients before and after chemotherapy and/or surgery. To evaluate the prognostic value of serum sCD44v6 and HA in patients with oral and maxillofacial malignancy and its value to monitor treatment response in these patients.1. Study of Levels of serum sCD44v6 in patients with oral and maxillofacial malignancyMethods: 28 patients, hospitalized from 2000 to 2001 with oral and maxillofacial malignancy were analyzed and 28 healthy individuals were also studied as controls. The patients aged from 29 to 80 with a mean of 55.32. The lesions included were az follows: oral and maxillofacial squamous cell carcinoma(OSCC) 17 cases, salivary adenoid cystic carcinoma(ACC) 6 cases, and others 5 cases. 18patients in stage I and II, 10 in stage III and IV. Venous blood were collected from these patients before therapy(surgery and/or chemotherapy) and the healthy individuals. And one week after therapy, venous blood were collected in 24 patients once again. Serum samples were obtained by centrifugation(3000rpm) and stored at -80 until assayed. Serum levels of sCD44v6 were measured with quantitative Enzyme-linked Immunosorbent Assay(ELISA) with commercially available kits. The test were performed according to manufacturer instructions. Serum levels of sCD44v6 were expressed in ng/ml. The student's test was used to assess the significance of difference in levels of serum sCD44v6 between two groups of samples.Results: Before initiation of therapy, no difference was noted between the sCD44v6 serum concentration in patients with oral and maxillofacial malignancy and control group(p>0.05). Levels of serum sCD44v6 in patients with OSCC and with salivary ACC had no difference with control group respectively (p>0.05). The sCD44v6 serum concentration of patients in stage IIL IV with oral and maxillofacial malignancy was higher than that of patients in stage I II and that of the control group, but the difference was not significant(p>0.05). Serum sCD44v6 levels in patients with OSCC after a complete treatment became lower than before treatment(p<0.05).Conclusion: Serum levels of sCD44v6 may not beuseful to diagnosis of oral and maxillofacial malignancy, however, it may be contributory to monitoring treatment response in patients with OSCC .2. Study of Levels of serum HA in patients with oral and maxillofacial malignancyMethods: 44 patients with oral and maxillofacial malignancy were analyzed and 24 healthy individuals were also studied as controls. All of them were the admitted patients for surgery or chemotherapy from 2000 to 2001 in Oral Department (aged from 45 to 80 with a mean of 60.32, including oral and maxillofacial squamous cell carcinoma(OSCC) in 34 patients, salivary adenoid cystic carcinoma(ACC) in 6 patients and other type's malignancy in 4; 31 patients in stage I and II, 13 in stage III and IV). Venous blood were collected from these healthy individuals and the patients before meripy(surgery and/or chemotherapy). And one week after therapy, venous blood were collected in 20 patients once again. Serum samples were obtained by centrifugation(3000rpm) and stored at -80 癈 until assayed. Serum levels of HA were measured with quantitative radioimmunoassay (RIA) with commercially available kits. All tests were performed according to manufacturer instructions. Serum levels of HA were expressed in ng/ml. The student's test was used to assess the significance of difference in levels of serum HA between two groups of samples.Results: Before initiation of therapy, the HA serum concentration in patients with oral and maxillofacial malignancy was markedly higher than that of the controls(P<0.05).
