Histomorphological Study On The Effect Of Hyaluronic Acid On Oral Wound Healing In Rats

Objective:In this experiment,0.2% hyaluronic acid gel was applied locally to the soft tissue wound of the hard palate of the rat to make a preliminary observation and to observe the pathological changes of the soft tissue wound of the oral hard palate,and to explore the effect of exogenous hyaluronic acid on the healing process of the oral wound.Methods:Forty-eight healthy SD male rats were randomly divided into two groups,experimental group and blank control group,24 rats in each group.Each experimental rat was made into a circular full-thickness defect with a diameter of 3 mm from the midpoint of the hard palate first molar to the distal third molar.Experimental group: Hyaluronic acid gel was applied to the wound on the day of surgery,and the drug was administered on the 1st and 2nd days after surgery,once a day.The blank control group was not treated after the hemostasis on the day of surgery.On the 1st and 2nd day after surgery,the sterilized injection water was administered for treatment once a day.Six animals were sacrificed on the 3rd,7th,10 th,and 14 th day after surgery.The mandible was separated and photographed under the same parameters.The image was imported into Image-Pro Plus 6.0 to measure the wound area,and the average wound surface area was calculated.Peeling the whole layer of hard palate mucosa,histological observation and histological standard quantitative evaluation after HE staining.Results:1.General observationIn the experiment,the oral hard palate wounds of the two groups of rats gradually healed with time.On the third day after operation,it was found that the bone surface of the wound was covered with a layer of serum cellulose,and the edge was red and swollen.The edge of the experimental group was more prominent than the control group.On the7 th day after surgery,the edge of the wound became irregular and the edge moved to the center.The area of the experimental group was smaller than that of the control group.On the 10 th day after operation,the wounds of the two groups were covered by the epithelium and the center was depressed.The experimental group had a larger depression than the control group.Fourteen days after surgery,the wounds in both groups were covered with epithelium and the scar was contracted.2.Analysis of mean wound surface area(MWSA)Comparison between groups: The mean wound area difference between the two groups on the 3rd,7th and 10 th day after surgery was statistically significant(P<0.05),and the control group was higher than the experimental group.There was no significant difference in wound area(P>0.05).Comparison of different time points in the group: The difference of the time points in the experimental group was statistically significant(F=1399.107,P=0.000),and the results of the two comparisons were available.The third day after surgery> Day 7> Day10> On day 14;the difference in the control group at different time points was statistically significant(F=477.752,P=0.000),and the results of the pairwise comparison were available.On the 3rd day,7th day>10th day>14th day after surgery.3.HE staining resultsOn the 3rd day after operation,no epithelial tissue was formed in the wounds of both groups,and inflammatory cells infiltrated.The experimental group had more inflammatory infiltration than the control group.On the 7th day after surgery,some newborn epithelium was seen in the wounds of both groups,and the inflammatory cell infiltration was alleviated.In the experimental group,collagen incorporation was observed;on the 10 th day after surgery,the intact epithelium was observed in theexperimental group,and the epithelium in the control group was incomplete.On the 14 th day after surgery,the intact epithelial structure appeared in both groups,and the inflammatory cell infiltration was significantly reduced.The keratinized layer was thicker than the control group.4.Quantitative evaluation of histological criteriaThere was no significant difference in the quantitative evaluation of histological criteria between the two groups on the third day after operation(P>0.05).There were differences in the quantitative evaluation of histological criteria between the two groups on the 7th,10 th and 14 th day after surgery.Statistical significance(P<0.05),and the experimental group was higher than the control group.Comparison within the group: There was a statistically significant difference at different time points in the experimental group(F=100.532,P=0.000),and the results of the pairwise comparison were available.The 14 th day after the operation> The 10 th day>The 7th day> The 3rd day There were significant differences in the control group at different time points(F=89.020,P=0.000),and the results of the two comparisons were available.The 14 th day after the operation> The 10 th day> The 7th day and the 3rd day.Conclusion:0.2% hyaluronic acid gel can promote wound healing in the early stage of wound healing,accelerate the healing process,and reduce the inflammatory reaction during healing and promote the regeneration of mucosal epithelium.
