Viral myocarditis (VMC) is a common disease that is caused by viruses,and its main histopathologic changes are mononuclear infiltration and myocardial necrosis. Year after year the morbidity of the VMC is rising and we always using the Chinese traditional medicine to therapy for viral myocarditis. In this experiment we compared the tumor necrosis factor titers of peripheral blood of ICR(Institute of Cancer Research) mice which induced by Coxsackie virus Bs(CVB3) on different days after infected between the control group and Shengmai treated groups. ICR mice were divided into five groups:uninfected group,virus-infected group and mice were injected daily with Shengmai injection starting on day -1 (the day of inoculated with virus is day 0),dayO,day+1 groups. The TNF-a titers of peripheral blood were measured by enzyme-linked immunosorbent assay (ELISA);the exist of CVBs-RNA in the myocardium were detected by In Situ Hybridization (ISH);The apoptosis were found in the myocardium by TDT/dUTP Nick End Labeling (T/UNEL);The changes of histopathology were observed under the light and electronic microscopy.The results were:(1) TNF-a titers were increased significantly in the infected mice,especially on the 5th day. (2) TNF-a titers were decreased significantly in the Shengmai treated group. (3) We found none of the mice had histopathologic changes and CVBaRNA in myocardium was negative in uninfected group,while mice in infected groups had varying degree of histopathologic changes. (4) The group of injecting with drug starting on day-1,the histopathologic changes was significantly less than the group of injecting with drug starting on dayO and day+l(P<0.05),and there was no different between the group of dayO and day+l(P>0.05). (5) The positive signal of apoptosis in myocardium was observed in the infected group.The results were suggested that TNF-a may play an important role in the early stage of VMC and in vivo it doesn't have anti-virus effects;Shengmai may prevent the early viral myocarditis through decrease TNF-a titers. In the therapy of virus induced disease drug prophylaxis is the most important,apoptosis is one of the mechanisms for increasing the myocarditis.
