It is well known that hcyatocellur carcinoma(HCC) is amalignan tUmor fOr its hyPervascullar, higher inVasive or metaStaticactivity and bad prognosis. The invasion and metaStaSis oftUmr cellis a comPlex Process. But the matrix metaloP rote inas es (MMP s)Prodced by W cell play a critcal role in metaStaSwi toremte organs and inVading W tissues, for MMPs candegrede extraCellular matrix (ECM) and basemen membrance whichthe toor cells must trgyerse. Therefore, it will be helPful tO5elucidate the maCanism of origin and metaStaSis of HCC tOinvestigate the reationshiP between MMPs and HCC. Since tissueinhibitOrs of metaloPrOteinase (res)can inhibit the aciVity ofMs,so it is imPOhallt tO study the oos in the HCC tissue. It wasroported that Matrix M etaI l OP rOte inas e- 9 (MMP -9 ), whi ch was on e o fMs boly, was associated with tUmor invasion particulary andcould be inhibited by tissue inhibitOr ofmetalloProteinase-l (TIMP-l).So it wil1 Wide theoretic basis for diagnosis, iudging the stage ofHCCs inVasion and predicting the Prognosis, finding new methd ofthN' There wee some rrportS abollt the relationshiP betweenM-9, M-1 and others thershu the commn view is notfoUnd at Presot .The relationshiP between or-9,TIMP-l and HCChad not been rrported. In the presCfl StUdy, we dctected the exPrssionof M-9, TIMP-1 by immUnohiStOchendmp Md in 42 samPlesof hcyatOcell carcinOma to investigate the relationshiP between theMMP-9, un-1 and HCC.Materials and methodsi The hssue of HCC was Obtained frOm42 patients who were underwen SUrgery .There were 33 males and 9femals.The age of patients ranged frOm l2 tO 76 years old (mean f 50.4years old ). 30 patens' AFP levels exceed 400ng/ml (76.6%). 29patientS' HBsAg was postive (68.9%). The diameter (d) of tumor6(d<5cm,9 samPles,dto5cm,33 samPles ) ranged frOm 2.0 cmtol7.5 cm (meanf8.l cm).TUmors wer gradd according tO thecriteria desribed by Edmondson and Steiner Grade I 3 samPles;GradeII 20 smples; Grade III 16 samPles; Grade IV 3samPles. 9 samPleswer well encaPsulated, 33 samPles were POOr encapsulatal, 12samPles with cancer emboli in the branchs of portal vein or hopaticveins. No patied have received radioativity, chemotheraPy, TACand irnmunothery before the SUrgery. The samPles taken from three cases of laceration injny of liverwere used as normal contrls.the POlyclone antibodes ofati-MMP-9, ami-TTMP-1 Wer USedto perfOrmed otohistochemical staining (SABC methd ).The irnrnunhistochemical staining of MMP-9 and TIMP-l isassessed according to both the distribuon (the percentae of POslerecells )and the intensity ofstaning. The statistical analysis was exeC'Ute by SPSS softwar. P valuesless than 0.05 are considered significen.ResultS f (1) The exPrssions of MMP-9,oc-l in normalcOIltrls were all negatiVe. (2)The positive rate of MMP-9 andW-l in HCC were 73.8 % (31/42) and69.0 %(29A2) resPectively.7The positive fate of MMP-9 and TIMP-l in the tissue near HCC is24.3%(8/33) and 36.4%(l2/33) reSPectiVely. There are obviousdiffeence ho the positive fate of MMP-9 and TIMP-l betWeenHCC and the tissue near HCC resPectively. (3)There were notobvious relationshiPs bCtween MMP-9 and AFP, HBsAg, age grous, sex (P>0.05).So does TIMP-l. (4An the grOUP of well differeniatied(I+II), the positive rate of MMP-9 is 65.2%,the POOdr differeniatiedM (III+IV) was 84.2%,There was no obVious diHce betweenthemre>0.05).(5) The exPression of MMP-9 was related to POorencaPsuating, porta embolus the0.05, P<0.05), bu TIMP-l was norelated to them ry>0.05).(6) In higher inVasiVe and metaStatic grou, the posihve nde of MMP-9 was 9l.0%, the lower invasive andmetaStatic grou was 55.0%, and was signfican differencesMcally (P<0.05).The positive of TIMP-l in the tWo above grOuPswas 86.3%, 50.0% resPectiVely, there is a significandiffereneto<0.05).(7the the HCC, There wer 6 cases with posihvestaining of M-9 and negative staining of TIM...
