Objective: To investigate expression of matrix metalloproteinase-1(MMP-1) and tissue inhibitor of metalloproteinases-1 (TIMP-1) in esophageal squmous carcinoma and the expression and the biological behavior of esophageal squmous carcinoma. Methods: Esophageal tumour samples(n=36) were obtained from esophagectomy specimens from patients(age range 35-70 years;29 males and 7 females) who had undergone surgery for esophageal cancer and none had receiced chemotherapy or radiotherapy.All specimens which had been submitted to the Department of Pathology in The First Affiliated Hospital of Jiang Xi Medical College(time range March 2000-November 2001).There are all squmous carcinoma.Among the 36 cases,6 tumors were well-differentiated squamous cell carcimomas,15 were moderately differentiated and 15 were poorly differdntiated;14 cases with lymph node metastases,22 cases with lymph node nometastases;11 cases invased to submucosal carcinomas and muscularis propria,25 cases invased to esophageal adventitia.The expression of MMP-1 and TIMP-1 of 36 cases of esophageal squmous carcinoma were detected by immunohistiochemistry(the method of ElivisionTM plus). Results: 1 ,Immunoreactivity of MMP-1 is found evenly in the tumor cells,while rarely in the interstitial cell.The expressive rate of MMP-1 of esophageal squmous carcinoma was 38.8%,and the expressive rate of MMP-1 was independent of tumor differentiation(P>0.05),higher rate of lymph node metastasis(P<0.05),higher rate of cases invased to esophageal adventitia(P<0.05). 2,Immunoreactivity of TIMP-1 is found evenly in the tumor cells and interstitial cell. The expressive rate of TIMP-1 of esophageal squmous carcinoma was 50.2%, and the expressive rate of TIMP-1 was independent of tumor differentiation(P>0.05),lower rate of lymph node metastasis(P<0.05),lower rate of cases invased to esophageal adventitia(P<0.05). 3,The cases which the expression of MMP-1 preponderated over the expression of TIMP-1 easilier occurred lymph node metastasis than the cases which the expression of TIMP-1 preponderated over the expression of MMP-1. Conclusions: 1,The expression of MMP-1 and TIMP-1 has a relationship with the biological behavior of esophageal squmous carcinoma.The positive control for MMP-1,while the negative control for TIMP-1. 2 ,The Blance MMP-TIMP which keep integrality and internal environment of extracellular matrix was determinant,and provided important target spot for the therapy of tumor.
