Objective:To study local pathological changes of tumor and the effects on the body immunological function of VX2 tumor rabbits after RFA treatment, and offer some experimental evidences of the application of RFA for entity tumor therapentics . In addition, the aim is to verify immune enhancement reaction of the body to the stimulation of tumor tissues which were retained in the body after hyperthemmy solidifying , which is so called the effect of tumor vaccine , so as to offer some experimental evidences to study the mechanism of action of tumor vaccine after hyperthemmy solidifying .Methods:1 .Tumor tissues in the effective range were gained immediately after the RFA treatment, and fixed by formaldehyde , stained by HE , and then were observed pathological changes under light microscope ;2. Rabbit VX2 cancer in liver was treated with RFA by the help of laparotomy , the activity and IL-2 secretion of T lymphocyte cells of rabbit blood , and the activity of macrophages in the abdominal cavity were measured regularly by MTT colorimetric assay .3.The rabbits transplanted VX2 tumors in the muscles of hind legs were divide into two groups , then were treated with operation resection and RFA respectively . All the rabbits were reinoculated with tumor after three weeks , and observed the growth conditions of the reinoculation tumor in the subsequent four weeks , then were killed , and the both lungs tissues of the rabbits were taken to count metastasis tumors by compressed tablets counting method.Results:1.The changes of tumor tissues could be observed with light microscopeafter RFA treatment: the cancer cells showed coagulation necrosis , intercellular space increased size , the intercellular substance disappeared , the cell body showed pycnosis , nucleus showed concintration and malformation , and the staining deepen compared to the pretherapy . The plasm proportion increased size which showed eosinophil dyeing , and the normal tissue showed no significant damages .l.Contrasted with control and pretreatment groups , the activity and IL-2 secretion of T Lymphocytes cells and the activity of macrophages in cavitas abdominis of radio-frequency ablation(RFA) treatment group were improved significantly (P<0.05) .3.The success rate of reinoculation tumor of the rabbits treated with RFA(18.75%) was significant different from the other (87.50%) (P<0.05) ; and reinoculation tumor of the rabbits treated with RFA grew obviously more slowly than the other group . Tumor metastasis rate of the rabbits treated with RFA (25.00%) was significant different from the other(93.75%) (P<0.05) , and the number of the metastasis tumor in the rabbits treated with RFA was 4 +2 , but the other was 10+6 .Conclusions:The tumor tissues in the valid range can be changed into coagulation necrosis tissues by RFA , but the normal tissues show no significant physics lesion , the RFA treatment can achieve the same effect as surgery resection . The RFA treatment can relieve the tumor loading of the body , retract the immunosuppression condition , improve the cellular immunity of the immune system, and inhibit the metastasis, regenesis, growth of the remaining tumor in vivo and the survival of the reinoculation tumor tissues , so RFA is an effective tumor therapeutical tool. The mechanism of action and pathway of RFA , enhancing the organical immunity , which is uncertain at present , is expected to explore furtherly .
