Objective To explore the relationship of HLA-DM genetic polymorphisms with susceptibility , autoantibody and activity of systemic lupus erythematosus and to analyse environmental risk factors and interaction between environmental factors and HLA-DM gene in SLE.Methods Genotypes of HLA-DM were determined by PCR-RFLP. The relationship of HLA-DM genetic polymorphisms with susceptibility of SLE was analysed by case-control study and the relationship of HLA-DM genetic polymorphisms with autoantibody and activity of SLE was analysed by case-case study . The environmental risk factors of SLE were analysed by logistic regression and the interaction between environmental factors and HLA-DM gene in systemic lupus erythematosus was analysed by generalized linear model.Results 7 kinds of allele frequencies and 42 kinds of haplotype of HLA-DM were no significant different between SLEs and controls. Genotype frequency of DMB*0102/0103 tended to increase in controls, but the difference was not significant after corrected (P<0.05, Pc>0. 05). 7 kinds of allele frequencies of HLA-DM were no significant association with antinuclear antibody and activity in SLE. The frequency of DMB*0101 tended to increase in anti-dsDNA (-) group of SLE, but the difference was not significant after corrected (Pc>0. 05). The frequency of DMB*0103 tended to increase in anti-dsDNA (+) group of SLE(Pc<0.001) and OR was 6.49.5 environmental risk factors (damp, mind stimulation , sunlight, sword bean, infection of tonsil) were selected to enterthe logistic model .The analysis of generalized linear model cued to exist interaction between HLA-DMB*0102 and infection of tonsil. Conclusions These data suggest that neither the DMA nor the DMB genetic polymorphisms determines susceptibility to SLE in Chinese, but there are significant association between DMB*0103 and anti-dsDNA in SLE and the SLE patients of DMB*0103 allele tend to present anti-dsDNA (+). There are multi environmental risk factors in SLE and there are the interaction between environmental factors and HLA-DM gene in systemic lupus erythematosus.
