【Objective】 To investigate the synergistic effect of tumor necrosis factor-αand interferon-γon the proliferation of newborn rat Schwann cells in virto. 【Methods】 Newborn rat Schwann cells were cultured in virto. Then the cultured and purified Schwann cells were identified with immunohistochemistry and electron microscop. The percentage of purified Schwann cells was over 90%. Schwann cells were cultured in the medium containing different concentrations of TNF-α,IFN-γ or TNF-αand IFN-γfrom newborn Wister rats and incubated for 24,48,72,96h respectively. We used tetrazolium salt (MTT) colorimetric assay to study the effects of TNF-α,IFN-γ or TNF-αand IFN-γon Schwann cell growth. 【Results】 The proliferative responses of Schwann cells were directly inhibited by TNF-αor IFN-γ. TNF-αinhibited proliferation in dose and time dependent manner in the levels from 4000U/ml to 16000U/ml. The rat of inhibitory effect can reach maximum when the concentration in 10000U/ml at 3 days. However, A synergistic greatly increase in the inhibitory effects of TNF-αand IFN-γ. The rat of inhibitory effect can reach maximum with the respectively concentration in 2000U/ml at 2 days. 【Conclusions】 TNF-αinhibited Schwann cells proliferation and in dose and time dependent manner. A synergistic greatly increase in the inhibitory effects of TNF-αand IFN-γ.These results give experimental evidence for the multimodality therapy of human tumor in the clinic.
