Background: Interleukin-4 is a pleiotropic cytokine secreted by Th2 cells and mast cells. Along with IL-13, IL-4 is a central mediator of allergic inflammation when asthma is going. Both IL-4 and IL-13 utilize IL-4R alpha chain as a component of their cognate receptor complexes. So IL-4R alpha chain gene has been thought as one of candidate genes of asthma.Objective: The aim of this study is to detect the association between polymorphisms of the S786P and I50V in the interleukin-4 receptor alpha chain gene and asthma in Chinese Guangdong Han population.Methods : Genome DNA from leukocyte was extracted as PCR templet. The polymorphisms of S786P are identified by RFLP-PCR and the polymorphisms of I50V are documented by ARMS-PCR.Results:1 Detecting the site of S786P in exon 12 of IL-4R a geneOnly S786 exits in the Chinese Guangdong Han population, and P786 was no found in the Chinese Guangdong Han population.2 Detecting the site of L50V in exon 5 of IL-4R a geneThe research group has been divided in two groups. The frequency of A allele is 49.5% and G allele is 50.5% in 103 asthmatic people. The frequency of A allele is 47.6% and Gallele is 52.4% in 62 controls.Conclusion:1 There is no polymorphism for S786P in exon 12 of IL-4Ra gene , and it is not thought as a candidate gene in the Chinese Guangdong Han population .2 There is I50V polymorphism in exon 5 of IL-4Ra gene in Chinese Guangdong Han people.3 No difference has been found between the controls group and moderate and severe asthmatic group. There is no association between the I50V polymorphism of IL-4Ra and asthma.4 No statistically difference has been found between the controls and asthmatic people. There is no association between the I50V polymorphism of IL-4Ragene and asthma.
