Obejective Experimental models had been established by porcine serum intraperitoneal injection (PSII) and common bile duct ligation (CBDL) in rats,in order to investigate pathogenesis, pathophysiology, prevention and therapeutics for the hepatic fibrosis. Methods In a total of 200 male Wistar rats weigh of 100-150g.Porcine serum intraperitoneal injection (PSII) group(40 rats) were established by continuous intraperintoeal injection of porcine serum with 1 ml/time dosage (3times/week) for 56 days, and common bile duct ligation (CBDL) group(40 rats) by the double ligation of common bile duct for 56 days. Following investigating markers were used in different periods (14days. 28days, 42days, 84days) after the models induced(l).Body weight; (2). Liver and spleen weight; (3).Hisotopathology of the liver under both optical and electron microscope; (4). Mast cell count (MCC);and (5)The changes of the hydroxyproline concentrationof the liver. Results l.CBDL group showed a reduced body weight during the all periods in comparison with SPII and control groups (P<0.01) . There was no difference of the body weight between SPII and control groups (P>0.05); 2. At 14 days, the increased hepatic weight in CBDL group were shown than that in SPII group.At 28 days ,the increased splenic weight in SPII and CBDL groups were shown than that in controls (P<0.01); 3. Hepatic fibrosis occurred in both PSII and CBDL groups at 14 days, (P<0.05) succeeded model rate both were 100% and no death.The typical characterof liver cirrhosis occurred at 42 days in PSII groups and at 28 days CBDL groups. At 42 days , the death rate in PSII groups was zero and was 60% in CBDL groups.4. Both SPII and CBDL groups shown the increasing tendency of Mast Cell Count (MCC) than that in contols (P<0.05). At 14days,28days, the level of MCC was higher in CBDL than in SPII (P< 0.05), but less at 42days (P<0.05); 5. At 14 days, hydroxyproline concentrations in SPII and CBDL were higher than that in the controls. (P < 0.05)Hydroxyproline concentrations had no significant difference between SPII and CBDL groups.(P > 0.05),At 42days ,there was the higher hydroxyproline of hepatic concentration in CBDL group than that in PSII groups(P<0.05).Conclusion (1)Ideal practicable models for hepatic fibrosis could be obtained by both intraperitoneal injection with porcine serum and ligation of common bile duct.(2)The model was induced by intraperitoneal injection with porcine serum in favor of the observation in situ , and had significance in the clinical and experimental liver transplant in rats.(3)The model with hepatic fibrosis induced by ligation of common bile duct was suitable to the investigation of the obstructive jaundice and liver cirrhosis. (4)The Masson trichrome stain can be used for an important method for the observation of the fibrostic cells, The changes of the mast cell could be considered as an assist marker for the observation of the liver fibrogenesis.In addition the concentration of the hepatic hydroxyproline could be used for diagnosis of the hepatic fibrosis. clinical and experimental study.
