Objective: The acute cholecystitis caused by obstruction of common bile duct is a common disease. But there are a few reports about the acute cholecystitis caused by obstruction of common bile duct.There is no animal study about Interleukin-6 and gallbladder tissue Polymorphonuclear leukocyte when obstruction of common bile duct causes the acute cholecystitis in china.This reseach was to take different operations and observe ligation and release ligation different time serum IL-6,gallbladder tissue Interleukin-6 (IL-6) and Myeloperoxidase (MPO), gallbladder pathological,to offer the theory and experiment support for cure common bile duct ligation induce cholecystitis at clinic.Methods: 54 healthy rabbits weighing 2.2-2.5Kg were randomly assigned to 9 groups equally; The normal control group (N)(n=6), the group of common bile duct ligation for one day(L1) (n=6), the group of common bile duct ligation for three days(L3) (n=6), the group of common bile duct ligation for five days(L5) (n=6), the group of common bile duct ligation for seven days(L7) (n=6), relieving ligation after common bile duct ligation for one day(R1) (n=6), relieveing ligation after common bile duct ligation for three days(R3) (n=6), relieving ligation after common bile duct ligation for five days(R5) (n=6), relieving ligation after common bile duct ligation for seven days(R7) (n=6). All the rabbits were prohibited to eat but not prohibited to drink before operation. After taking different operations,the group of bile duct ligation rabbits were killed after ligating for one day, three days, five days, seven days and the relieving ligation group rabbits were killed after relieving ligation for three days. Getting 5 milliliter blood from every rabbits. Radioimmunoassay was used to measure Ilb -6 in serum and gallbladder tissue. Colorimetry were used to measure MPO in gallbladder tissue.The gallbladder pathologic changes,especially PMN in gallbladder tissue were observed with light microscope after HE staining. The experimental data were expressed as mean±standard deviation.The experimental data were analyzed by SPSS 13.0,a statistic sofeware.Results:Earlier period of common bile duct ligation(L1, L3 group), gallbladder intumesce, gallbladder paries, edema, incrassation, membrane plica integrated.Light microscope: membrane epidermis integrated, inherent membrane, muscle, serosa edema, blood vessel dilate, congestion, and a few PMN soakage. Along with ligating time, gallbladder inflammation aggravate ,floccule effused, paries become incrassation, bile thickness , membrane accrete ash yellow exudation or come into being sham film, part membrane putrescence desquamate, forming small ulceration. gallbladder nigrescence, paries become weak, membrane plica disappear, take on putrescene.Light microscope: membrane epidermis desquamate, blood vessel dilate, congestion, membrane, inherent membrane, muscle, serosa have a plenty of PMN soakage. L5 can be found gall-stone. membrane epidermis disappear, a plenty of white blood cell oddment and bleed, muscle layer slur, often concomitancy gallbladder around inflammation, when inflammation become severe.Gallbladder tissue IL-6 and MPO:Gallbladder tissue IL-6 and MPO gradually went up with ligating time prolong, and lifted up to top level in the group of bile duct ligation seven day(L7).Compared with every group of common bile duct ligation was significant difference(P<0.05). After relieving ligation,the levels of IL-6 and MPO in gallbladder tissue reduced significantly than the group of common bile duct ligation(P<0.05).Serum IL-6: Serum IL-6 gradually went up with ligating time prolong, and lifted up to top level in the group of bile duct ligation seven days(L7). The IL-6 of L5 went up significantly than the group of normal concrol group(P<0.05).After relieving ligation,serum IL-6 reduced significantly (P<0.05).Conclusions:1 IL-6 plays an important role in the happening of cholecystitis which was induced by common bile duct ligation. 2 Gallbladder tissue Polymorphonuclear leukocyte increases with ligating time prolong. Inflammation aggravates wirh it.Finally gallbladder tissue is damaged aggravatelly. Relieving ligation can lighten damage.3 Common bile duct ligation can induce cholecystitis.Along with ligation time prolong, gallbladder inflammation aggravates. After relieving ligation, gallbladder inflammation lightens. Relieving ligation earlier ,inflammation lighten faster.
