Objective: To compare the efficacy of oxibendazole (OBZ), pre-liposomal albendazole (Pre-L-ABZ) with albendazole Tablet (ABZ-T) and Lipsomalalbendazole (L-ABZ) against experimental Echinococcosis granulosis cyst in mice. Methods: The 148 mouse were developed for ten weeks after intraperitoneally inoculated and randomly allocated in groups as follows: (l)ABZ-T oral administration (Po- ABZ-T )with 75 mg/kg dosage;(2)OBZ oral administration with 37.5 mg/kg dosage; (3)OBZ oral administration with 75 mg/kg dosage; (4) OBZ oral administration with 150 mg/kg dosage; (5) Pre-L-ABZ intraperitoneal injection (Pi- Pre-L-ABZ )with 75 mg/kg dosage; (6) Pre-L-ABZ oral administration(Po- Pre-L-ABZ) with 75 mg/kg dosage; (7) L-ABZ intraperitoneal injection (Pi-L-ABZ )with 75 mg/kg dosage; (8)Saline oral administration with 0.3ml/per mouse dosage; 9. Saline intraperitoneal injection with 0.3ml/per mouse dosage; All groups were administered with 3 times/ week medication for 10 weeks; Following investigating markers were used after overall administration: 1. Mean cyst weight and its reduction rate after treatment; 2. Histopathological changes of the cyst under both optical and electron microscope; 3. Measurement of the concentrations of the OBZ, ABZ and its metabolites, ABZ-sulfone (ABZSN) and ABZ-sulfoxide ( ABZSF), in plasma, liver and lung tissues; Results: The mean cyst weights andreduction rates were 159.33 ± 263.21 (82%) ,49.75± 43.62 (95%) ,and 191.5 ± 98.36 (79%) in different OBZ dosages with 37.5mg/kg, 75mg/kg and 150mg/kg groups respectively; The mean cyst weights and reduction rates were 87.59±95.63(86%)and 58.0 ± 151.0(83%) in Pi-Pre-L-ABZ and Po- Pre-L-ABZ groups respectively; The mean cyst weights and reduction rates were 143.75 ± 106.62 (76%) and 145.13 ± 162.47(84%)in Pi-L-ABZ and Po-ABZ-T groups; The significant difference of mean cyst weight were seen between various drug groups and controls with saline oral administered and intraperitoneal injected groups (P < 0.05), but not between OBZ, Pre-L-ABZ, L-ABZ and Po-ABZ-T groups(P >0.05); Histopathological changes of Eg.cyst showed the significant difference between different dosages in OBZ oral groups and saline oral administered group (P<0.05), and Pre-L-ABZ, L-ABZ groups and saline groups (P<0.05), but there were no difference between the OBZ groups and ABZ-T group (P>0.05) , Pre-L-ABZ groups and ABZ-T, L-ABZ groups (P>0.05). Concentrations of ABZSN and ABZSX in plasma and liver exhibited about two times higher in Pre-L-ABZ groups than that in Po-ABZ-T group(P<0.05). Conclusions: 1. OBZ was highly effective against experimental infected echinococccsis granulosis cyst in mice, the efficacy of OBZ is about to equal with ABZ-T; 2. Pre-liposomal ablbendaole can be considered as an effective anti-hydatid drug, may have a significant role of passive targeting liver tissue, it showed about 2 times higher ABZSN and ABZSX concentrations in the plasma, liver tissue than that in Po-ABZ-T group in mice.
