Renal carcinoma is one of malignant tumors with high incidence rate. It is the tumor with second incidence rate in urinary surgery and only inferior to tumor of bladder. Its biological behaviors are very mul-tiform, and the pathogenesis and molecular biological basis of the tumor are not clear. PKC is one kind of protein kinase located in cyto-plasm, and distribute in organ, tissue and cell of the body widely. The kinase has very important physiological function. PKC play multi-ple roles in cell physiological regulation, such as induce cellular growth, perform information convection, regulate the expression and function of nuclear gene, and affect cellular cycle and apoptosis. At present, 12 kinds of PKC isoforms have been dissociated and purified, and they are coded by different genes. Their biochemical structures are different, and appear obvious specificity of histocyte in the func-tion. Lots of research have indicated PKC has close relationship with tumor. In the paper, the differences of expressions of different PKC isoforms in different staging and grade renal carcinoma and normal re-nal tissue have been examined and the relationship between PKC iso-forms and renal carcinoma has been analyzed. We hope the results can supply theory basis to promote therapy of renal carcinoma.Experimental materialThe 30 specimens in the experiment came from renal carcinomaand normal peripheral tissue resected by operation in our hospital from 2000 2001. Fresh tumor tissue and normal renal tissue 3cm far from tumor were selected, and the specimens would be sent into refrigerato-ry with -70?C. A group of cases ( aged from 37 to 77, averaged 57. 0, 21 male and 9 female) was involved in the experiment. The renal lesions included 16 left renal carcinomas and 14 right renal carcino-mas , in which 25 accepted radical operation of renal carcinoma by ab-domen and 4 did radical operation by waist. All cases were primary and didnt perform radiotherapy, chemotherapy and immunotherapy. Pathological staging referred to Robson staging method, and histologi-cal grade referred to Cleveland Clinic grade criteria.Experimental methodAgents and method: histone III - S, protamine, first antibody of PKC isoform a and e, and second antibody of goat anti - rabbit anti-body were from Sigma Company; [ γ ?32p ] supplied by Furei Com-pany. Extracted cytoplasm, protein of membrane of all specimens, performed protein quantu with Folin - phenol agent. Applied Western Blotting method to examine the expression of PKC isoforms ( PKC - α, PKC ?e) in renal carcinoma tissue and normal tissue.Statistical analysis; quanto data was analyzed by X2 test with SSPS statistical software.Experimental results1. By pathological staging, the specimens in the experiment were divided into 4 groups including 7 cases with I stage, 18 cases with II stage, 4 with III stage and 1 with IV stage. By histological grade, the specimens were divided into 4 groups including 6 cases with I grade,17 cases with fl grade, 5 with 1Hgrade and 4with Fgrade.2. The relationship of the expression of PKC isoforms between re-nal carcinoma and normal tissue:The expressions of PKC isoforms in renal carcinoma and normal tissue were obvious different. Compared with normal tissue, the ex-pression of PKC - a in cytoplasm of renal carcinoma significantly de-creased; the expression in the membrane obvious increased, and the differences had statistical meaning { p < 0. 01). Compared with nor-mal tissue, the expression of PKC - e in cytoplasm and membrane of renal carcinoma significantly increased, the differences had statistical meaning (p <0.01).3. Trie relationship between PKC isoforms and staging and grade of renal carcinoma tissue:When performed analysis, we put IV tumor into III group, and did 4 grade tumor into 3 grade group. There was relationship between the expression of PKC - a in renal carcinoma tissue and staging and grade of tumor.
