| The study is to isolate and purify chondroitin sulfate from pig cartilage and investigate the effect of anticoagulation of chondroitin sulfate and the influences'of acetaldehyde and ethanol upon this effect. The method of dilute-enzyme hydrolysis has been used to extract chondroitin sulfate from pig cartilage. Next, the salt in the crude product has been get rid of by using gel filtration chromatography. The comparative experiment shows that Sephe-dax G-25 has advantage over Sephedax G-100 in purify effect. The purification process has been done by ion exchange chromatography. After comparision of three ion exchange resins and three dilution acidities, the optimum dilution condition has been determined. In the course of identification, the results show that the product has no protein, no peptide, no amino acid and on other glycosaminoglcans by using Lowry's test, thin section chromatography and cellulose acetate membrane electrophoresis. The infrared scanning characteristics of the product is in conformity with that of Sigma's standard sample. The purity of the product is 99. 7472% by the examination of high-performance liquid chromatography (HPLC). All these results show that the product can be regarded as chromatopurity. The study also evaluated the anticoagulation function of chondroitin sulfate by examining clotting time(CT) and found that chondroitin sulfate has certain anticoagulation effect(14. 33 ± 0. 72, P<0. 01) , but much lower than that of heparin(87. 04?. 20). By preincubated with 500mmol/L acetaldehyde, the clotting time of chondroitin sulfate has been prolonged (27.59±3. 59), while 500mmol/L ethanol has effect upon both chondroitin sulfate and heparin(P>0. 05), which indicate that the acetaldehyde can enhance the anticoagulation effect of chondroitin sulfate, but ethanol has no this effect.
