Objective To investigate the effect of regulation of qi and the blood drugs (RD) and compound prescription(RP) on melanocytes(MC) and tyrosinase activity(TC) in vitro, and discuss its therapeutic mechanism for pigmented diseases.Methods Human MC were cultured in vitro and treated with 4 concentrations of the RD and RP ranged from low to high:0.001g/L, 0.01g/L,0.1g/L,lg/L. Eight drugs were divided into five groups: wind-dissipating and blood-moving(WB), qi-moving and depression-resolving(QD), blood-quickening and stasis-transforming (BS),Liver-supplementing and kidney-enriching(LK) and compound prescription (RP), empty was given to contrast group. The morphologic changes of MC were observed in comparison with that of the control group. The proliferation of MC, melanin synthesis and tyrosinase activity were determined by MTT assay, Hunt method and oxidation of DL-dopa respectively.Results WB:With the concertrations of Angelicae Dahuricae Radix at O.Olg/L and 0.1 g/L, The proliferation of MC and melanin synthesis were inhibited, However, they were inhibited by low but activated by high concentrations of Tribulus terrestris L. At the same time, TC were inhibited by low but activated by high concentrations of the both(P<0.05). QDrThere were no significant effects on the 3 indexes after treated with Curcumae Tuber and Cyperi Rhizoma ( P>0.05 ). BS: After being treated by Angelica sinensis(Oliv.) Diels and Ligustici Rhizoma at the dote of 0.0001 g/L to Ig/L, MC proliferation, melanin synthesis and tyrosinase activity were increased significantly. Moreover, the activation effects of Angelica sinensis(Oliv.) Diels were in a concentration-dependant manner(P<0.01). LK:The 3 indexes were increased significantly by the activation of Lycii Fructus and Hominis Placanta at the dose of 0.001 g/L to Ig/L, and the activation effects of Lycii Fructus were in a concentration-dependant manner too(P<0.01). RP: RP had a strong promotion on melanocytic proliferation, melanin synthesis, and tyrosinase activity at four concentrations comparing with other groups(P<0.01).Conclusion Regulation of qi and the blood drugs increased melanocytic proliferation, melanin synthesis and tyrosinase activity, and the test level was thecompound prescription for complex effect, of which might be the mechanism that made it valid in clinical treatment of the pigmented skin diseases.
