| ObjectiveTo study the effect of curcumin on the expression of the relative activity of melanocytes,tyrosinase, melanin synthesis, melanocyte migration and c-kit mRNA content.MethodsMedium with different concentrations of curcumin in vitro melanocytes were detected usingThe cell activity was determined by MTS assay after culture for24hã€48h; NaOH pyrolysismethod and Oxidation dopamine response in vitro is used to detect melanin synthesis andtyrosinase activity; Cell scratch experiment to observe the melanocyte migration situation; Annexin V-FITC staining was measured cell apoptosisapoptosis; Real-time fluorescentquantitative PCR detect c-kit mRNA’s relative expression.Result1. At different concentrations of curcumin(5.0μmol/lã€10μmol/lã€20μmol/l) showed no significantimpact of the human epidermal melanocytes compared to the control group (all p>0.05), whenintervented the melanocytes for24h,but the concentrations of30μmol/l of curcumin can inhibitthe proliferation of melanocytes,the difference are significant(p<0.05); when cultured48h,Curcumin concentrations of less than20μmol/l, have no significant compared to the controlgroup(all p>0.05), the30μmol/l of curcumin can inhibit the proliferation of melanocytes, thedifference are significant (P <0.05).2. The30μmol/l of curcumin can induce cell apoptosis, the apoptosis rate was10.02%at24h,the apoptosis rate was11.70%at48h, there is a significant difference compared with the controlgroup (both P <0.01).3. Curcumin concentration of less than20μmol/l can inhibit the synthesis of melanin andtyrosinase activity, compared with the control group, the difference are significant, whenintervented the melanocytes for48h (all P <0.05).4. Curcumin concentration of less than20μmol/l can inhibit the cell migration compared with thecontrol group, the difference are significant, when intervented the melanocytes for48h (all p<0.05).5. Real-time fluorescent quantitative PCR showed that different concentrations of curcumin(5.0μmol/l,10μmol/l,20μmol/l) can suppress the expression of c-Kit mRNA compared with thecontrol group (all p<0.05).Conclusion1. A low concentration of curcumin have no effect on melanocytes, high concentration ofcurcumin have cytotoxicity for melanocytes.2Curcumin(≤20μmol/l)can inhibit the melanin synthesis, tyrosinase activity, migration ofmelanocytes.3Curcumin (≤20μmol/l)can reduce relative expression of human epidermal melanocytes ofc-kit mRNA. |
