Objective To study the effects of Dex and TMPz on the expression of c-fos,bcl-2,ICAM-1 protein after rats renal ischemia-reperfusion at different time point.Methods 150 male Wister(weight was 250±15g)rats were randomly divided into five groups: the sham-operated (sham)group,ischemia-reperfusion(IR)group,Dexamethasone (Dex+IR)group,Tetramethylpyrazine (TMPz+IR )group and Dex+TMPz+IR group. Every group had thirty rats,and every time point there were six rats under examination after reperfusion for 1h,3h,6h,12h,24h. Immunohistochemical and HE staining were used to examine the positive expression rate of c-fos,bcl-2 and the microvessel count of the positive expression of ICAM-1 protein. Immunohistochemical parameters were compared within groups and chi-square test,Variance analysis were used for statistic analysis.Results Localization of c-fos protein expression was found in nuclei of the proximal convoluted tubules,distal convoluted tubules and collecting duct cells. c-fos protein expression started to increase at 1h,peaked at 3h,reduced rapidly after6h.Bcl-2 mainly located at the cytoplasm of proximal convoluted tubules ,protein expression was increased at 1h after reperfusion, peaked at 6h,maintained to 24h.ICAM-1 protein located at microvessels and tubules of renal, the expression of ICAM-1 started to increase at 1h,peaked at 24h.After injected Dexamethsone and TMPz, the expression of c-fos and ICAM-1 protein decreased significantly (p<0.01), however, bcl-2 increased significantly(p<0.01). Compared with TMPz+IR group and Dex+IR group, the c-fos and bcl-2expression of Dex+TMPz+IR group were quite different at the several time points(p<0.05).Conclutions The apoptosis of renal after ischemia-reperfusion injury may be activated by increasing c-fos,ICAM-1 protein expression and inhibited by increasing bcl-2 protein expression. Dexamethasone plus TMPz may decrease c-fos,ICAM-1 protein expression and greatly increase bcl-2 protein better than using the two medicines separately.
