Objectives:To explore the possible protective effect and the related mechanism of Sinomenine on acute renal ischemia reperfusion injury in rats models.Methods:A renal ischemia reperfusion model according to John D Hughes was produced by 30 male SD rats,which were randomly divided into 5 groups:(1)Control group,which include 6 rats,just removal the right kidney;(2)Renal ischemia reperfusion injury groups(IRI group),which include 12 rats, and it divide into two goups due to the examine time after reperfusion,these rats'right renal will be removed and these left renal artery will be occlusion by small clips for 45 minutes; (3)The sinomenine precondiontioning groups(SIN group).which also include 12 rats,The animals were treated as same as IRI goup except Intraperitoneally by SIN 80mg/kg before surgery,in mean time Control goup rats and IRI goup rats should Intraperitoneally by saline Equivalent as SIN goup. All animals will be killed in right time after blood samples were gathered and left renal were cut for exanination. the levels of serum creatinine(Scr),blood urea nitrogen(BUN),serumβ2-microgobulin(β2-M);serum and renal tissue's super oxide dimutase(SOD), mlondialdehyde(MDA);renal tissue's expression of intercellular adhesion molecule-1(ICAM-1)in each group were respectively detected after renal ischemia reperfusion for 0 minutes and 4 hours,Pathological changes were also examined by light microscope.Results:(1) Also there were statistic difference between SIN group and Control group,sinomenine precondiontioning decreased the levels of Scr,BUN andβ2-M significantly (P<0.05). (2) There were statistic difference of the serum levels of SOD,MDA and renal tissue's SOD,MDA in sinomenine precondiontioning group and IRI group ( P < 0.05 ) .(3) the score of renal tubular injury in sinomenine precondiontioning group reduced significantly compared with IRI group (P<0.05).(4) Sinomenine precondiontioning decreased the index of ICAM-1 expression and the score of renal tubular injury significantly(P<0.05). Also, they had a significant positive correlation with each other(r=0.888,P=0.000).Conclusions:(1)Sinomenine precondiontioning can protect the kidney against renal ischemia reperfusion injury.(2) Sinomenine can reduce the expression of ICAM-1. and may protect the kidney against renal ischemia reperfusion injury by the effect of Inflammatory and Antioxidant.
