Study Of Ziqian Anticancer Preparation

According to the multiplex anticancer mechanism and considering traditional Chinese medicine principles,ZIQIAN anticancer preparation is a new complex one which contained with anticancer activity compounds in materias. The formulation is composed of Radixa Arnebia seu Lithospermi, Radix Rubiae, Rhizoma zedoariae and Herba Eishoitziae seu Moslae.In the prescription,Radixa Arnebia seu Lithospermi,Radix Rubiae,Rhizoma zedoariae as well as Herba Eishoitziae seu Moslae is monarch ministerial adjuvant and conductant drug respectively. In the experiment　we got the optimal extraction technique through measuring and comparing the content of effective components in the half-finished product and product and discussed the methodology of quality control of ZIQIAN anticancer preparation. we also screened the optical matching components in the prescription on tumor model used as mice bearing sarcoma 180 and identified the extraction and preparation by TLC. The effective components were measured according to pharmacopeia. It was demonstrated that those contents in the materials used in our experiment were comply with pharmacopeia . The optimal extraction technique of Radixa Arnebia seu Lithospermi,Radix Rubiae was screened with Uniform experimental design method,as the contents of naphthoquinone and anthraquinone were used as marker. The results wasshown that the optimal extraction technique as follows: Radixa Arnebia seu Lithospermi was soked with 11 times that amount of 95% ethanol at the temperature of 30℃ for 3 hours,twice and the herb of Radix Rubiae was refluxed with 10 times that amount of 70% ethanol at the temperature of 100℃for 4.5 hours ,twice.According to literatures Rhizoma zedoariae and Herba Eishoitziae seu Moslae were extracted through the optimal technique and the contents was measured respectively. Chemical compounds in Zedoray oil were identified qualitatively by means of GS-MS. Simultaneously the optimal condition of preparing Zedoray oil withβ-cyclodextrin inclusion was discussed through Uniform experimental design method. It was proved that the optimal condition was to use 5 times of β-cyclodextrin than Zedoray oil to include with stirring for 3 hours used with magnetic stirrer at the temperature of 30℃. We screened the optimal matching components in prescription with the tumor model used as mice bearing sarcoma 180 and observed its inhibition ratio of tumor growth .On the basis,the ZIQIAN capsule was made, which contained Radixa Arnebia seu Lithospermi and Radix Rubiae extraction 1.98g, 0.5g respectively and the oil of Rhizoma zedoariae 0.32g,Herba Eishoitziae seu Moslae extraceion 0.13g in a daily dose. The qualitative differentiation of the extractions of Radixa Arnebia seu Lithospermi,Radix Rubiae and preparation was studied. The contents of levo-shikonin and rubimaillin in the herb of Radixa Arnebia seu LithospermiandRadix Rubiae and their extractions as well as in the preparation were measured and compared differentially andqualitatively.It was proved that the extraction technique is reasonable and method of the research for the quality control of ZIQIAN anticancer preparation is also reliable.
