The Expression And Significance Of Pgp,MRP₁,LRP In Transitional Cell Carcinoma Of The Bladder

Objective To study the expression and significance of Pgp MRP1 and LRP in transitional cell carcinoma of the bladder(TCCB).Methods The immunohistochemical staining with S-P method is used to examine the Pgp MRP1 and LRP expression in 65 cases of primary TCCB,15 cases of recurrent TCCB and 12 cases of normal bladder tissue.Results l.The expression rate of Pgp MRP1 and LRP in the 12 cases of normal bladder tissue is 41.7% (5/12) 16.7%(2/12) and 33.3%(4/12) respectively;that in the 65 cases of primary TCCB is 72.3% (46/65) 70.8%(46/65) and 76.9%(50/65) respectively;and that in the 15 cases of recurrent TCCB is 93.3% (14/15), 86.7%(13/15) and 93.3%(14/15) respectively.The expression of Pgp MRP1 and LRP in the recurrent TCCB is significantly higher than that in the primary TCCB respectively (P<0.02;0.02 and 0.02),and the latter is significantly higher than that in the normal bladder tissue(P<0.05;0.002 and 0.01).2.Both of the Pgp and LRP expression in the primary TCCB have positive correlation with the pathological grade of TCCB(P=0.0007 and0.0451),and a negative relationship is observed between the MRP1 expression and the tumor grade (P=0.0197).There is no significant difference between the superficial primary TCCB and the advanced primary TCCB for Pgp MRP1 and LRP expression respectively (P>0.05).The expression of Pgp MRP1 and LRP correlates well with the results of anti-cancer drug sensitive assay respectively (P<0.05).There is no correlation among the expression of Pgp MRP1 and LRP in the primary TCCB(P>0.05).Conclusions Pgp MRP1 and LRP are significantly higher expressed in the primary and recurrent TCCB and all the three proteins play important roles in the primary and secondary multidrug resistance of TCCB. The high expression of Pgp LRP and low expression of MRP1 may be marks of the loss of cell differentiation for primary TCCB.All the three proteins have good correlation with the results of anti-cancer drug sensitive assay.Detecting the expression of Pgp MRP1 and LRP simultaneously will help select anti-cancer drugs in clinics.
