Along with the rapid progress of the kidney transplantation,the organ preservation solution need improvement. It should be not only effective but also convenient for blending,storage and transportation in the future,and moreover it may be put in some immunosuppressive agents with gene-engineering techonology,such as IL-10 CTLA-4Ig,TGF- P Fasl et al which can conduce the preserved organ to acquire immunity tolerance .Consequently,the postoperative rejection rate would decrease .Our research project will be divided into two steps .Firstly,develop a kind of organ preservation solution which is in accordance with the situation of our country,cheep and reliable convenient for blending -. storage and transportation .Secondly,add gene-related therapentic factors to organ preservation solution . The first step is going on .Having analyzed the compenent of the UW and HTK organ preservation solution thoroughly and take the advantage of Chinese traditional medicine,we pull off the following integrant menu Major integrant of multi-organ preservation solutionPH:7.45+0.10 Osmolarity(mmol/kg):320-340 Characteristics of this kind organ preservation solution are following:l.Histidine/ Histidine monohydrochloride replace Lactobionate,which isnon-osmotio integrant can suppress the tissue acidification effectively under so high density as 180/18(mmol/L).2.Dex-40 replace Hydroxyethyl starch. Varapamil is capable of suppressing the inner cellular calcium over-loading,abating the calcium-pump loading of membreme of cell,mitochondria and endoplasmic reticulum during cold storage,then reduce energy consumption,and consequently the energy balance remain in a long time.The calcium regulation is not solitary,but related with the regulation of Na+ and K+.Even though during normal temperature,ion regulation need energy as well,during cold storage,ion regulation consumpt much more energy because of worse ion disorder.If ion channel blockers is used to lower the transmembreme potential,cell energy would be saved.TEA Quinidine and Amiloride suppress Na+-Ca2+ Na-K Na-H exchangingrespectively,and work together to prevent cells from swelling caused by Na+ K+ disorder.Furthermore TEA also suppress Na-Ca2+ exchangeing,lighten Ca2+ loading and as a result economise energy.During low temperature,abnormal exchangeing of K+ and Na+ leads cell swelling,and Ca2+ over-loading give rise to cell damage.If the ion channel blocks were used to lighten ion disorder,the energy consumption would go down,and in a beneficial circle,energy storage is conductive to ion balance.The successful organ preservation solution should possesses the following traits:(1)abate cell swelling (2)avoid cell acidification (3)avoid enlarging the space between cells (4)lighten reperfusion injury (5)supply material to synthesize energy substance.
