HC-A preservation solution has been used in the field of kidney transplantation in our country for over twenty years and has a positive effect. Along with the rapid progress of the kidney transplantation and the development of Ischemia/Reperfusion theory ,the organ preservation solution need improvement.. Having analyzed the compenent of the UW and HTK organ preservation solution thoroughly and take the advantage of Chinese traditional medicine,we pull off the following integrant menu.According to the clinical experience,we have found drawbacks of HC-A preservation solution as following: a high concentration of Magnesium Sulfate makes deposition in the preservation solution and renal vessels during a cold circumstance, Mannitol may be harmful to kidney, pH of HC-A preservation solution is too low and unstable which can not prevent the acid of renal cells.Characteristics of HCA-II preservation solution are following:1. The concentration of Magnesium Sulfate is reduced to suppress it's deposition in the preservation solution and renal vessels .2.Histidine/ Histidine monohydrochloride replace Mannitol, which is non-osmotio integrant can suppress the tissue acidification effectively under so high density as 160/16(mmol/L).3.Adding Dex-40 can enhance preservation effect.lt has been found that the bill secreting function of isolated liver preserved for 48 hours remains normal in the group in which Dex-40 replace Hydroxyethyl starch,but decrease by 65 percent in the contrast group(UW group). It is believed that DEX-40 is able to improve micro-circulation and benefit to organ reperfusion.4. Ligusticum Wallichii entract agent(Ligustrazine) is added on. Ligustrazine contains SOD which can clear away oxygen free radical,flavonoid glycoside compound which can remove oxygen free radical directly,and terpeneester which reduce the production of oxygen free radical by blocking the PAF acceptor.As of potential to capture OH-factor and the effective integremt of Ligustrazine is equivalent to hydromedin which is well known as oxygen free radical scavenger and calcium ion blocker.The successful organ preservation solution should possesses the following traits:(l)abate cell swelling (2)avoid cell acidification (3)avoid enlarging the space between cells (4)lighten reperfusion injury (5)supply material to synthesize energy substance.We evaluate the effect of this kind organ preservation solution with preservation and transplantation of dog's kidney.In preservation test, kidney underwent histologicaK biochemical analysis and energy substance PH measure at different phrase of preservation. In transplantation test,we transplantated dog's kidney by the support of related research unit.The result of the experiment show that the HCA-II preservation solution reachs on the whole the five sorts of need above mentioned,and the surviving rate of transplanted organ was satisfactory. In conclusion,the HCA-II preservation solution we researched and developed is not only effective,but low cost convenient for blending storage and transportation,therefore it accords with the situation of our country.
