Objectives:1. To investigate the expression of a novel cell adhesion molecule co mplex which is composed of trophinin,tastin,and bystin in human endometrium,and its relationship with some sorts of infertility.2. To detect the expression of trophinin in human embryo and its characters.3. To study the role of the complex on human placentation and the influence of Mifepristone on the expression of the complex in human villous trophoblast.Methods:1. Immunohistochemistry technique with specific monoclonal antibody was applied to investigate the relationship between a novel cell adhesion molecular complex which is composed of trophinin,tastin,and bystin and infertility by detecting the expression of the complex in normal cycling endometrium and the differentces between the normal and the infertility.2. IIP technique with specific monoclonal antibody was applied to investigate the expression of trophynin in human blastomere and blastocyst,which was gained with the sequential culture in vitro.3. Immunohistochemistry technique with specific monoclonal antibody was applied to detect the expression of the complex in human placenta villi of different phases of pregnancy,meanwhile the difference of the expression of trophinin,tastin and bystin in human placenta from 14 sugical abortions by negative pressure suction at clinics (as control) and12 incompletely drug abortions by dilation and curettage (as experimental cases) were also studied.Results:1. Trophinin,tastin,and bystin were expressed in the luteal-phase endometrium,and the strongest in the mid-luteal phase(P<0.001). Compared with normal women,immunostaining of trophinin,tastin in the mid-luteal endometrium with endometriosis,unexplained infertility was significantly weaker (P<0.05).2. The expression of trophinin was increased with the development of human embryo,and the polarity of the molecular expression was perceived in human blastocysts acquired with sequential culture in vitro(P<0.05).3. Trophinin,tastin,bystin in controls were expressed in a stronger way in the cytoplasm of syncytiotrophoblast,About 10 week of pregnancy,the expression was very weak(P<0.01),and after 12th week of pregnancy,the complex were not detected in placenta villi. The expression of trophinin and bystin in syncytiotrophoblast of the experiment group I decreased significantly compared to the control group(P<0.05),and so did the expression of tastin in syntiotrophoblast at the same time.Conclusions:1. Trophinin,tastin,bystin expressed regularly in human endometrium,which was the strongest in mid-luteal phase,compatible with the opening of embryo implantation window,may play important roles in the process of embryo implantation. Abnomal endometrial trophinin,tastin,andbystin expression might be one of the major causes of infertility.2. Trophinin expressed in human blastocyst is a special molecule mediating a unique homophilic cell adhesion between trophoblast and endometrial epithelial cells at their respective apical cell surfaces,and the homophilic adhesion regarded as a new acting mode between blastocyst and endometrium maybe an important mechnism in embryo implantation.3. It can be inferred that the adhesion molecule complex likely to devote itself to the placenta formation according to the special feature of the complex expression. Mifepristone acted strong influence on the expression of trophinin,tastin,bystin in tropoblast,which may be one of the target tissues for Mifepristone,and the complex may have relation to the process of abortion.
