Objective: To study the significance and expression of p27 , p53 and PCNA in endometrial Hyperplasia and endometrial adenocarcinoma.Methods: p27,p53 and PCNA expression in paraffin-embedded sections of 33 endometrial adenocarcinoma, 20 simple endometrial hyperplasia, 16 complex hyperplasia and 20 atypical hyperplasia were detected by immunohistochemical s-p assay.Results: The rate of the high immunostaining of p27 in simple endometrial hyperplasia, complex hyperplasia, atypical hyperplasia and endometrical adenocarcinoma were 75%, 56.3%, 35% and 30%, respectively. There was a significant difference between endometrial adenocarcinoma and endometrial hyperplasia (p<0.01). The expression of p27 was not correlated with histological grade, muscles invasion or patient's age(p>0.05). The positive immunostaining rates of p53 in atypical endometrial hyperplasia and endometrial adenocarcinoma were 15% and 54.6%, but no p53 expression was observed in simple and complex endometrial hyperplasia. Significant difference of p53 expression was found between endometrial hyperplasia and endometrial adenocarcinoma. No correlation was found between p27 and p53 expression in endometrial adenocarcinoma. Intense expression of PCNA increased from simple and complex hyperplasia to atypical hyperplasia and endometrial adenocarcinoma. An inverse correlation was found between p27 and PCNA expression in endometrial hyperplasia(p<0.01). An positive correlation was found between p53 and PCNA expression in endometrial hyperplasia and endometrial adenocarcinoma (p<0.05).Conclusion: Reduced or absent p27 expression may play an important role in the genesis of endometrial adenocarcinoma . The protein expression of p53 may play an import role in the genesis and development of endometrial adenocarcinoma . Endometrial adenocarcinoman is related both to the abnormal regulation of cell cycle and to the augmentation of proliferative activity in endometrial hyperplasia and endometrial adenocarcinoma. Comprehensive analysis of p27, p53 and PCNA expression could help to understand the mechanism on endometrial progression to malignancy...
