Objective To explore the mechamism of treating leukemia with Yiqi Yangyin formula. Methods We hatched cell lines K562 with human leukemia by means of serological pharmacology. At the same time, Yiqi Yangyin formula is given to the rats. And then, we observe rats' serum's effects on membrane fluidity, cell cycle and bcl-2 gene and mutant p53 gene expression of cell lines K562 by fluorophotometry, flow cytometry and immunohistochemic. Results Serum of the rats given Yiqi Yangyin formula at lower drug concentrations has little effect on the membrane fluidity. But at higher drug concentrations it can increase membrane fluidity of cell lines K562, this effect has the same tendency to serum of the rats given arsenical agents and that given HU. Serum of the rats given Yiqi Yangyin formula can increase membrane fluidity and S phase cells' percentage of cell lines K.562 and decrease GO-G1 phase cells' percentage, which is similar to the effect of the rats' serum that are given arsenical agents. Serum of the rats given Yiqi Yangyin formula down-regulates the expression of bcl-2 gene and mutant p53 gene, and in addition it induces the apoptosis of the cell lines K562. Conclusion With serum of the rats given Yiqi Yangyin formula, S phase cells of cell lines K562 increased, and then apoptotic cells increased with a diminution of S phase cells. This mechanism may relate to the function that increase membrane fluidity of cell lines K562 and down-regulate the expression of bcl-2 and mutant p53 gene.
