BACKGROUNDEndometriosis(EMS) is a common gynecologic disease, me incidence rate is increased per year gradually, and become the hot spot of gynecology in recent years. The mechanism of pathology is not clear to this day, but attachment, aggression, angiogenesis has become the generally accepted"AAA"program of the implantation and grow of endometrium. Cell adhesion molecule has a closely relationship with this procedure, it is the glycoprotein of the cell surface, including immunoglobulin super family, integrin family, selectin family, cadherin family et al. Integrin avb3 has many functions which including adhesion of cell to extracellular matrix, platelet aggregation, immunal function, tissue repair, tumor infiltration, angiogenesis and the acceptance of uterus. So to study the role of integrin 33 in EMS is significance to the pathogenesis of EMS. The metastasis of tumour cell is related with active integrin 33, at present, the antibody of anti- has been used to treatment of tumour metastasis on abroad, while the cell of endometriosis is similar to the tumour at the biology behavior. So use the antibody of anti 33 to therapy the endometriosis, maybe have the new world to the treatment of endometriosis.The recurrence rate of endometriosis is high after the drug treatment and operation. So people study the new and effective method to treat theendometriosis up to now. Because of ethics problems, it is difficult to study in human body, especially it is impossible to practice the injury examination on human body repeatedly. Therefore to study the pathogenesis and therapeutic efficacy, it is necessary to establish the model of endometriosis. It is an ideal pathway to establish model of endometriosis on Rhesus macaque, but because of the animal is too expensive and deficiency of the animal, this way is restricted. SCID is severe combined immunodeficient mouse, it is rarely occurrence reject reaction when implantation the foreign tissue on the body of the mouse.. The cost of mouse is not expensive and the source of animal is plenty. So SCID is an ideal animal to establish the model of endometriosis. We establish the animal model of endometriosis on SCID successfully, and have a good base for the pathogenesis and treatment of endometriosis.OBJECTIVEStudy the expression of integrin 33 in eutopic endometrium, ectopic endometrium and normal endometrium, to investigate the mechanism between the integrin 33 and endometriosis rudimently. Establish the implantation animal model of endometriosis, to investigate the relationship of aggression and metastasis between the activation of integrin 33 and endometriosis. which provided an themry base for the further investigation of the etiology and medical therapy of EMS.METHODS AND RESULTS1. Culture the cell of eutopic, ectopic and normal endometrium: the endometriumwas digested by the collagenase then cultured.2.The expression of integrin 33 on eutopic, ectopic and normal endometrium: We use the flow cytometry to detect the expression of 33 on the 13 case of eutopic endometrium, 12 case of ectopic endometrium and 10 case of normal endometrium.3.The establishment of animal model of endometriosis: The eutopic and ectopic endometrium was implanted into the subcutaneous of abdomen of SCID mice, which was taken out at 8 weeks after the implantation and keep the serum to detect the marker. The focus which implanted into the subcutaneous of the mice, which was decreased size at first week because of the fluid of disposing the specimen was absorption, then the size increased gradually, but slowing. The size of focus between of 0.4-0.8cm. The tissue was made pathology examination. The section of tissue was staining with HE, which proved the good grow condition of the endometrium, the intact gland and stromal was detected. Use the immunohistochemical to detect the expression of cytokeratin, CD34 and integrin 33.
