Objective: The purpose of the study was to evaluate the effect of compound Salvia Unguent (Co-S-Ung) in inhibition of scar formation, to creat a cicatric animal model though repairing method of the lip cleft of rabbit and to investigate the pathologic changes during wound healing. Methods: Thirty New Zealand white rabbits were formed scar models by Millard' s method of cleft lip repair, in addition, one white rabbit was executed as control model. The experimental rabbits were divided randomly into three groups, 10 rabbits each group. The first group was treated with Co-S-Ung (experimental group), the second with menstruum unguent (menstruum group) and the third was treated nothing (control group). The volume of scar and granulation tissues were measured at 7,15, 21, 30, 60, 90 days postoperatively. The scaring tissue and granulation were harvested at 7, 30, 60, 90 days. The light microscopes and electronic microscopes on fibroblast, mast cell, myofibroblast and collagen bundles observed the pathologic changes of scar.Results. (1)The surface of scar was observed by naked eyes. The scar was swollen at 24 h postoperatively. The wound healed on 7d postoperative. The scar was contracted and stiffens on 2 Id postoperatively, soften and redden 30 days postoperatively. The scar of experimental group was more plane than that of another two groups 60d and 90d postoperatively. (2)Measurement of the scar: the volume of scar of the experimental group was significantly larger than which of the monstrous group and control group(P<0.01) 2Id to 90d postoperative.(3) Histopathological examination revealed the amount of fibroblasts and mast cells in experimental group were more than that of another two groups at 30d, 60d, 90d postoperatively. We observed that the fibroblasts of experimental group were rare, the rough endoplasmic reticulum was scarce, the nucleolus presented irradiation and the nuclear chromation were condensed and gathered to the side of nuclear membranes accompanied by many apoptotic body appeared. In another two groups there were many immature mast cells that released granulation andcontacted with fibroblast, the scars formed with a diffuse mass of irregularly arranged thick collagen fibers.Conclusion: The Co-S-Ung can reduce volume of scar, inhibit fibroblast proliferation, influence collagen protein synthesis and interfere mast cell releasing grain.
