Psoralea corylifolia L. is one of the commonly used traditional Chinese medicines included by the Pharmacopoeia of the People's Republic of China. Its seeds are known for their tonic, antibiotic effect and antiviral effect and so on. Coumarin and flavonids is the main component. 13 Samples of Psoralea corylifolia L. were collected and the quality assessment was developed. Conditions for the extraction of Psoralea corylifolia L. were studied by orthogonal experimental design. The effect of Psoralea corylifolia L. on the proliferation of UMR106 cell was studied. And the pharmacokinetics was studied in animals.1. Studies on the assessment of Psoralea corylifolia L.HPLC-FPS of different Psoralea corylifolia L. from different origins was studied comparatively. The results showed that this method had a good repeatability, and the ratio of common peaks' area of different samples were different. The method developed can evaluate the quality of Psoralea corylifolia L. conveniently.ABSTRACTA method for simultaneous determination of Psoralen and Isopsoralen in Psoraleae corylifalia.L by HPLC with UV detection is established. Bifonazole was used as internal standard. The detection wavelength was set at 245nm. These components were separated on a Hypersil ODS column with acetonitrile-H20 (30:70, v/v) as mobile phase. The linear ranges were 0.50 ~10.0|ag/ml for Psoralen and Isopsoralen. The method was proposed to be used for quality control of Psoraleae corylifalia L. and good results were obtained. The method is simple and accurate.2. Studies on the orthogonal experiments of Psoraleae corylifalia L.To optimize the conditions for the extraction of Psoralea corylifalia.Conditions for the extraction were studied by orthogonal experimental design as guided by the content of total flavonoids, Psoralen and Isopsoralen present in the extract. Significant effect was observed in concentration of alcohol for the total flavonoids in Psoraleae corylifalia L. The optimum condition for the total flavonoids in Psoralea corylifolia L. was A3B2C2, that is, adding ten times amount of 95% alcohol and refluxing and extracting for 2h(totally extracting for 3 times). The optimum condition for the Psoralen and Isopsoralen in Psoralea corylifolia L. was AjBiCi, that is, adding five times amount of 65% alcohol and refluxing and extracting for lh(totally extracting for 3 times).3. The Pearson correlation coefficients between some variable and total amount of flavonoids of Psoralea corylifolia L was founded and good results were obtained.4.Studies on the pharmacokinetics of Psoralen and Isopsoralen in rabbitsThe sensitive and specific HPLC methods were developed for determination of Psoralen and Isopsoralen in rabbit plasma, and the pharmacokinetics were studied in rabbits.After ig of Psoralea corylifolia L. extract, the plasma samples were extracted with n-hexane:ethyl acetate(13:l, v/v). The chloramphemicol was used as internal standard. The Psoralen and Isopsoralen were separated on a Hypersil ODS column with acetonitrile-HzO (30:70, v/v) as mobile phase.The methods were simple, accurate, sensitive and good enough to be used in pharmacokinetic study of Psoralea corylifolia L.
