[Objective] To observe the effect of ambroxol on nitric oxide synthase in rats withCOPD and discuss the mechanism of the preventive and therapeutic effect of ambroxol on COPD.(Methods] 32 male Wistar rats were randomly divided into three groups.Thenormal group (A,n=8) ,The model group (B,n=12) and the ambroxol treated gro up (C,n=12) .Ratsin group B and group C were established by intratracheal instill ation of lipopolysaccharide twice and exposed to cigarette smoke.The group C wa s treated with ambroxol sincethe first day.Pulmonary and bronchial morphological changes were observed by light and electron microscopy .The quantitative patholo gical examination by image analysis wasalso performed.The expression of inducibl e NOS (iNOS) and endothelial NOS (eNOS) inpulmonary tissue was observe d by immunohistochemistry technique.(Results] Compared with group A, group B presented significant differences in PaCO2. PaO2. MLI and MAN [ (53±4) mmHg vs (41 ±3.5) mmHg, (80. 5±8.6) mmHg vs (98.4 ± 2.9) mmHg, (92±10) urn vs (52±3) um, (268 ± 15 ) /mm2 vs (410±27) /mm2 (P<0.01) ]; The expression of eNOS and iNOS were also markedly elevated (P<0.01). In comparison with group B , group C present ed significant differences in blood gas analysis [PCO, (48±3mmHg vs (53±4) mmHg, P02 (89.2 ± 7.3) mmHg vs (80.5 ± 8.6) mmHg) (P<0.01)]. MLK MAN were significantly different[ (83 ± 8) urn vs (92 ± 10) urn (P<0.05) , (291 ± 10 ) /mm2 vs (268±15) /mm2 (P<0.01) .The expression of eNOS were decreased (P<0.01), while the expression of iNOS were increased (P<0.01) .[Conclusion] Smoking can induce over expression of iNOS and weak expre ssion of eNOS in pulmonary tissue.The former might result in lung injury and COPD.
