Objective: The objective of this paper is to do some research on the function of previous inflammation cytokines (PIC) and IgG subclass in children's asthma.Method: This paper mainly uses ELISA method to test the level of IL-6, IL-8 and TNF- a in serum and supernatant of peripherial blood mononuclearcyte ( PBMC) and concentration of IgG subclasses in serum in different periods of asthma (35 attacks, 18 stable condition). Results: The results illustrate that during the children's attacks, the value of serum IL-6, IL-8, TNF- a is obviously higher than that in the children's stable condition or in their normal period With the improvement of children's conditions, the amount of these three cykotines decreases quickly to or near the level of the normal group. The result of external experiments shows that during the children's spasmodic asthma period the level of IL-6, IL-8 and TNF- a , whether the PBMC is stimulated by PHA or not, goes up more prominently than that in the normal group. But compared with the normal group, the level of IL-8, TNF- a in the stable period is still higher weather the PBMC is stimulated by PHA or not. During the spasmodic asthma period the deficiency rate of IgG subclass is 28,57% (10/35), out of which the deficiency of IgG2 is most obvious (accounting for 61. 53%). At the same time, the increasing rate of IgG subclass is 97?4% (34/35), out of which IgG4 is its main factor (accounting for 94,28). But during the stable period, the rate of IgG subclass and (or) its deficiency degree and categories are all reduced. The value of serum IgG4 is directly proportional to that of IL-6, IL-8 and the same is true with the relationship between serum IgG4 and serum IL-8 and TNF - , out of which the latter is produced from PBMC in external experiment. Conclusion : The paper comes to the conclusion that there exists the imbalance of IgG subclass and the imbalance of PIC in the aspect of IL-6, IL-8 and TNF- . This may be the basis of molecular immunology in asthma. The most effective treatments are continuous cmti-infiammation and immunological regulation.
