Experimental Study Of The Yifuning Soft Capsules On Treatment Of Climacteric Syndrome

Objective: To study the pharmacodynamics on treating climacteric syndrome (CS) of traditional Chinese medicine formula Yifuning soft capsules (YFN), which can nourish the kidney and activate blood, and explore its therapeutic mechanisms by studying its effects on receptor-neuroendocrine-immune network. Method: We established the models of climacteric syndrome with ovariectomized (OVX) rats. After giving drugs 4 weeks, we detected serum sex hormone (E2 P, Te, FSH, LH), interleukin-2 (IL-2) and hypothalamic, pituitary and plasma P-endorphin ( P-EP) levels by radioimmunoassay. The estrogen receptor (ER) levels of uterus and spleen were detected with method of radioligand binding assay of receptor (REAR) .The hypothalamic monoamine transmitters (NE, DA, 5-HT, 5-HIAA) contents were detected by fluorospectrophotometric method. The adrenal gland, uterus, thymus and spleen index were calculated. Then adrenal, uterus and thymus pathologic changes were investigated with light microscope. Result: YFN can improve levels of serum , IL-2 and estrogen receptor of uterus and spleen in OVX rats and increase their adrenal gland and uterus index. YEN can increase reduced hypothalamic, pituitary and plasma P -EP levels in OVX rats. YFN can reduce contents of hypothalamic 5-HT and 5-HIAA, increase the levels of NE and DA in OVX rats and reduce the ratio of 5-HT to NE and 5-HT to DA in OVX rats. YFN can ameliorate the pathologic changes of the adrenal gland, thymus and uterus in OVX rats too. Conclusion: YFN may produce effects on declined neuroendocrine-immune network in menopause from various levels, ways and links by meansof regulating the function of plant nerve and reproduction endocrine and improving levels of ER and body immunity. So YFN can stabilize inner condition of body and relieve the syndrome of CS.
