Objective:To establish experimental pulmonary embolism model of animals in pregnancy for next research, no-pregnancy rabbits and pregnancy rabbits were injected self-blood clotting. As most contractive substance on vessel and trachea heretofore, endothelins-1 play an important role in many pulmonary disease. But we did not search the literature about relation of endothelins-1 between pulmonary embolism. To present the base for next research, endothelins-1 in blood serum and expression of endothelins-1 and pathogenesis in lung were detected in all rabbits.Methods:1. rabbits were divided into 4 groups. Rabbis of group A is pregnancy rabbits injected self-blood clotting into right atrium by femoral vein ;Rabbits of group B is pregnancy rabbits injected saline; Rabbits of group C is female no-pregnancy rabbits injected self-blood clotting into right atrium by femoral vein; Rabbits of group D is female no-pregnancy rabbits injected saline.2.Change of respiration and heart rate and survive ability of each rabbit were detected in each group.3.Each rabbit were exsanguinated by femoral vein before injected, instant injected, and 2 hour,4 hour,6 hour, and 8 hour after injected. Endothelins-1 in blood serum were detected.4.Change of endothelins-1 in lung were observed by method of immunohistoehemistry in group A and group B.5.Expression of mRNA of endothelins-1 in lung were detected by RT-PCR in each group.Results:1.A11 rabbits in group A showed symptom of pulmonary embolism andits mortality is 3/20. All rabbits in group B did not show symptom of pulmonary embolism and had not pathologic change of pulmonary embolism and die. All rabbits in group C showed symptom of pulmonary embolism and its mortality is 7/20. All rabbits in group D did not show symptom of pulmonary embolism and had not pathologic change of pulmonary embolism and die.2. Endothelins-1 increased in group A after embolism from instantly to 8 hour comparing with group B. Peak of Endothelins-1 in blood serum was in 2 hour after pulmonary embolism in group A. Endothelins-1 increased on group C after embolism from instantly to 8 hour comparing with group D. Peak of Endothelins-1 in blood serum was on instantly after pulmonary embolism in group C.3.There were expression of endothelins positive cell in lung in group A comparing with group B by staining of immunohistochemistry.4.mRNA of endothelins-1 of lung in group A and C expressed in high level comparing with group B and D.Conclusion:1.A11 rabbits by injected self-blood clotting showed serious symptoms of pulmonary embolism and its mortality is high. Pathological change of thrombotic pulmonary embolism in lung existed in group A and C.2. Endothelins-1 increased in group of pulmonary embolism after embolism from instantly to 8 hour comparing with group of no-pulmonary embolism.3.Expression of endothelins-1 increased in lung-the most important organ of producing endothelins-1.4.mRNA of endothelins-1 of lung expressed in high level while occurring pulmonary embolism.
