Objective: To measure the levels of p16 and cyclinD1 expression to investigate the genetic mechanism of uterine sarcomas and to produce the clue for the earlier diagnose and therapy of them. Methods: 28 uterine sarcoma tissues obtained including 12 uterine leiomyosarcomas and 7 endometrial interstitial sarcomas and 9 mixed mullerian sarcomas had been conformed by specialist in pathology. We made use of immunohistochemical staining S-P method to measure the levels of p16 and cyclinD1 expression in every uterine sarcoma at the against of 15 leiomyomas which is at the same time as 28 uterine sarcomas.Results: 1.The percentage of cyclinD1 expression positive and stongly positive cases was 46.4%,40% respectively and 100% was almost weak positive against p16 except a stongly positive case in uterine leiomyomas; the percentage of p16 and cyclinD1 positive was 67.9% and 60.7% respectively in uterine sarcomas.There was no difference (p>0.05) between uterine sarcomas and leiomyomas to cyclinD1 expression. However,there was significant change to p16 expression (P<0.05) betweenuterine sarcomas and leiomyomas . 2.The percentage of p16 positive cases was 66.7% in Leiomyosarcomas,71.4% in endometrial interstitial sarcomas and 66.7% in mullerian sarcomas.And cyclinD1 was 58.3% in Leiomyosarcomas,57.1% in endometrial interstitial sarcomas and 66.7% in mullerian sarcomas.When they were compared with each other,there was no significant difference (p>0.05). When compared with uterine leiomyomas, there was significant difference only for the expression of p16 in leiomyosarcomas(p=0.028). 3. the expression of p16 and cyclinD1 had no close corelationship in uterine sarcomas (rs′=-0.152,P=0.44). 4. the percentage of p16 positive of stage I-II and stage III -IV was 70% and 42.9% respectively in uterine sarcomas.And cyclinD1 was 70% and 71.4% There was no significant change (P>0.05)about the expression of p16 and cyclinD1 between stage I-II and stage III -IV uterine sarcomas .The phenomenon maybe result from a few cases. Conclusion: 1.The stongly expression of cyclinD1 may be an eanly event in the genesis of uterine sarcomas and general phenominone in loss of control during cell reproduction. 2.The abnormal expression of p16 may be implicated in the mechanism of uterine sarcomas progression and be considered as the difference mark between uterine sarcomas and leiomyomas. 3.The expression of p16 and cyclinD1 altogether dicovered the complex of genicexpression during the genesis of uterine sarcomas. 4. the expression of p16 wasn't closely related with cyclinD1,as showed that the absence of p16 and The stongly expression of cyclinD1 altogether or alone took part in the genesis of uterine sarcomas and suggested that there maybe exist other gene participating in the genesis of uterine sarcomas...
