Objective:The study is to investigate the expression and significance of P-STAT3 and cyclinD1 in the TMA of cervical tissues and analyze their relationship with generation,growth and metastasis function in cervical carcinomas,trying to search for the possible theory for diagnosis,prognosis and treatment of cervical squamous cell carcinoma.Methods:The expression of P-STAT3 and cyclinD1 proteins in the normal cervical squamous epithelium(NE),cervical intraepithelial neoplasia(CIN)and cervical squamous cell carcinoma was detected respectively by tissue microaway technique and SP immunohistochemical method and their relationship with clinical patho-parameter was analyzed.All data were decomposed by SPSS version 11.5 analysis software.Results:1.The result of experiment in tissue microarray(TMA):all chips were arranged orderly and clearly in the block after immunohistochemical staining.there were no shift,shrinkage and distort and meet to reseach.2.The positive rate of P-STAT3 were 67.5%in cervical squamous cell carcinoma,were 20.0%in CIN and were nonpositive in normal group.The differences were significant(P<0.05).3.The expression of P-STAT3 in different histological grades and clinical stages were various.The positive rate of P-STAT3 in gradeⅡandⅢof cervical carcinoma(60.3%)was much higher than that of gradeⅠ(10.0%),and the positive rate of P-STAT3 in stageⅢof cervical carcinoma(51.5%)was significantly higher than in stageⅡ(28.6%).The differences were significant (P<0.05).The positive rate of P-STAT3 was closely related to lymph node metastasis(P<0.05). 4.The positive rate of cyclinD1 were 72.5%in cervical squamous cell carcinoma,were 20.0%in CIN and were nonpositive in normal group.The differences were significant(P<0.05).5.The expression of cyclinD1 in different histological grades and clinical stages were various.The positive rate of cyclinD1 in gradeⅡandⅢof cervical carcinoma(80.4%)was much higher than that of gradeⅠ(20.0%),and the positive rate of cyclinD1 in stageⅢof cervical carcinoma(78.8%)was significantly higher than in stageⅡ(28.6%).The differences were significant (P<0.05).The positive rate of cyclinD1 was closely related to lymph node metastasis(P<0.05).6.There was significantly positive association between P-STAT3 and cyclinD1 expression(r=0.484,P<0.05).Conclusion:1.The expression of P-STAT3 and cyclinD1 in cervical squamous cell carcinoma was significantly higher than cervical intraepithelial neoplasia and normal cervical tissue.Both of their expression had a significant relationship with tumoe cell differentiated degree,clinical stage and lymph node metastasis.P-STAT3 and cyclinD1 may play a certain role in generation,growth and metastasis function in cervical carcinomas.2.There was a significant positive correlation between the expression of P-STAT3 and cyclinD1 in cervical squamous cell carcinoma,suggesting that STAT3 play an important role in the occurrence and progression of cervical carcinoma by increasing the expression of cyclinD1 directively or indirectively and Once Stat3 was activated,it could promote the expression of cyclinD1 and induce cell proliferation.
