Objectives :Osteoporosis(OP) is a disease characterized by low bone mass, deteriorated micro-architecture, hence resulting in bone fragility and following increasing risk of fracture. According to DEXA results in connection with population census in 2000 , population with primary osteoporosis account for approximately 6.97% of our country total population(about 88,000,000). All of women over 60 and men over 75 suffer from osteoporosis on different degree. Osteoporosis have been one of most severity problems threatening senile healthy. The rate of postmenopausal women suffering from osteoporosis is the highest—over 50% . The major cause is the standard of estrogen decreased rapidly which can rise rapidly bone resorption and lead to osteoporosis. Ovariectomized rat model displays many characteristics in common with postmenopausal osteoporosis, and has been widely used in the study of osteoporosis mechanism following estrogen withdrawal, and evaluating the therapy medicines. During screening and observing medicine , it remains an important issue—remedial time. Many researchers think that the obvious change on bone need three months at least due to slowly bone metabolizing. Bone modeling and remodeling include two major process: osteoblastic(OB) bone formation and osteoclastic(OC) bone resorption. OB and OC collaborate to change bone organic and inorganic matrix, bone histomorphology and BMD. We can shorten time of screening medicines and lessen workload through observing the change of OB and OC function, organic and inorganic matrix, bone histomorphology and BMD to find if medicines play role in preventing and treating osteoporosis.Currently, western medicine treat OP usually adopt HRT to control higher turnover bone metabolism and bisphosphonates to inhibit bone resorption. But estrogen can enhance the risk of breast and womb cancer and ALN has side effect on digestive tract, and they are restricted to clinically apply. So many researchers focus on searching effectively curative medicine from natural plant.Traditional Chinese medicine considers the kidney is the foundation of the native constitution, dominating bone, producing marrow, storing the essence, promoting reproduction. Menopause canresult in postmenopausal osteoporosis due to exhausting the essence, not satisfying bone. The spleen is the sea of food supplies, the source of producing gas and blood, the root of growth. In fact, senile people all have different spleen weakness. So postmenopausal osteoporosis have an affiliation with kidney and spleen. We choose Polygonum multifloruma to strengthen kidney, Astragalus mongholicus to enhance spleen and ALN, E2 ,GSK as control to treat OVX rat osteoporosis model in short-term. This study aims to supply short-term treatment evidence of screening therapy osteoporotic medicines and compare with pharmacological action of these medicines. Methods: 120 SD rat were divided randomly into 7 groups(16rats/group): sham ovariectomied group (SHAM), ovariectomied group (OVX), ovariectomied + Polygonum multifloruma group (HSW), ovariectomied + Astragalus mongholicus group (HQ), ovariectomied +E2 group (E2), ovariectomied + ALN group (ALN), ovariectomied +GSK (GSK) group. After 30days operation, began to treat these rats. When given medicine 15days and 30days, rats were killed seperately. After drawing blood from femur aorta the blood allowed to clot, we centrifuged and collected serum. We collected all femurs and tibias also. Serum and bone was for determination of biochemical marker, bone matrix,type I collagen by immunohistological method, bone histomorphology, BMD. It was observed if Polygonum multiflorum and Astragalus mongholicus effect on OVX OP rat during short -term treatment.Results:1 Results in First term (15days) treated OVXCompared with SHAM group, ALP, BGP, TRAP in OVX group had different degree increase both serum and bone.
