In this paper, the chemical constituents, quality control method and HPLC fingerprint chromatogram of Astragalus mongholicus were studied.The chemical constituents of Astragalus mongholicus were studied and 9 compounds were isolated and purified by silica gel, sephadex LH-20, and PHPLC column chromatography. Utilizing chemical and spectroscopic methods (UV, NMR, MS), the structures for total of 9 compounds were identified. These compounds belonged to flavonoids and saponins.A RP-HPLC was established for the simultaneous determination of six flavonoids in Astragalus mongholicus. The HPLC system consisting of a C18 column (250 mm x 4.6 mm id,5μm) and a mixture of acetonitrile and 0.05% phosphoric acid with gradient elution as the mobile phase was adopted. The column temperature was set at 35℃and the flow-rate was 1.0 mL-min-1. The absorbance was monitored at 210 nm and 250 nm. The linear response range was 3.312-33.12μg·mL-1(r= 0.9996),2.520-25.20μg·mL-1(r= 0.9992), 6.640-66.40μg·mL-1(r= 0.9992),10.80-108.0μg·mL-1(r= 0.9994),2.416-24.16μg·mL-1(r= 0.9993) and 2.790-27.90μg·mL-1(r= 0.9994), respectively (n= 6). The average recoveries (n= 9) of six flavonoids were greater than 95%, and relative standard deviations were less than 2.5%. The assay demonstrated that the method had adequate accuracy and selectivity to measure the concentrations of six flavonoids in Astragalus mongholicus.HPLC fingerprint chromatogram was established for Astragalus mongholicus. The HPLC system consisting of a C18 column (250 mm x 4.6 mm id,5μm) and a mixture of acetonitrile (A) and 0.05% phosphoric acid (B) with gradient elution (0-70 min,15%-40% A; 70-80 min,40%-40%A) as the mobile phase was adopted. The column temperature was set at 40℃and the flow-rate was 1.0 mL-min"1. The monitoring wavelength was at 203 nm. The HPLC chromatographic fingerprint of Astragalus mongholicus showing 17 characteristic peaks were established and 6 peaks were identified. The 19 batches of Astragalus mongholicus were classified to three groups based on the results of cluster analysis. The RP-HPLC fingerprint method is repeatable, feasible in analysis of Astragalus mongholicus and can be used scientifically to evaluate the quality of Astragalus mongholicus with quantitative assessment.
