| Object our past researches suggest ATRA plus bFGF would promote marrow stromal stem cells (MSCs) transdifferentiating into neuron-like cell. This paper study the effect and mechanism of bFGF on MSCs differentiating into neuron-like cells and action of c-myc in this process. It will offer important data for neuronal transplantation.Methods MSCs identified from adult human and rats were treated by different conditions: group 1: 2%DMSO+ 200umol/L BHA and serum freeï¼›group 2: 10ng/ml bFGF preinducted MSCs for a day , followed with 2% dimethyl sulfoxide (DMSO)+ 200umol/L butylated hydroxyanisole (BHA) and serum freeï¼›group 3:0.5umol/L all trans-retinoic acid (ATRA)+10ng/ml bFGF preinducted MSCs for a day, followed with 2%DMSO+ 200umol/L BHA and serum freeï¼›group 4: 10ng/ml bFGF pretreated MSCs for a day,followed with modified neuronal medium and serum freeï¼›Morphology of differentiated neuronal cells were observed, and the expression of neuron specific enolase(NSE),microtubule associated protein2 (MAP2),glial fibrillary acid protein(GFAP),NMDAR and 5-HT1D in differentiated neuron, the expression of FGFR1 and FGFR3 at pre and pro differentiation and the expression of c-myc during differentiation were detected by immunocytochemistry. Results After induction, MSCs transdifferentiated into a typical neuronal morphology at 1hr,, neuron-like cells expressed increasingly NSE,MAP2,NMDAR and 5-HT1D ,but didn't express GFAP at 24hr. There is a higher transdifferentiation percentage in the 2nd and 3rd group than that of the 1st group. Under 4th condition, almost 95% of the cells were switched toneuron-like cells. The expression of FGFR3, not FGFR1 was increased after differentiation. The protein of c-myc could be continually expressed from 4 hours to 36 hours during differentiation.Conclusion Different growth factors and chemical reagents can induce MSCs differentiated into neuron-like cells. There is a higher transdifferentiation percentage in the group pretreated by bFGF than that of the control.
