Objective To observe pathological alteration of relevant neurons in CNS after traumatic axonal injury for further exploring pathological mechanism of diffuse axonal injury (DAI) and providing experimental evidences for seeking and designing new target of treatment and improvement DAI. Methods A DAI animal model-optic nerve strech modal of rat was adopted. Stretch injury was induced in the optic nerves of adult Wistar rats. 50 Wistar rats were randomly divided into two groups. Then we chose different intervals (2h,8h,1d,3d,5d,7d) post-strech injury as the observational points. The conditions of tissue and cell injury were observed under microscopy and electromicroscopy. The neuronal injury was observed by TUNEL stain. Results Histological change included the loss of neurons and Vacuolation, electromicroscopic examination showed neuronal apoptosis and got the most serious 5 days after injury. TUNEL stain revealed positive from 1 to 3 days after injuryand reached the peak after 5 days. Conclusion Diffuse axonal injury can lead to delayed neuronal death, mainly exhibits apoptosis and a long duration of apoptosis.
